OBJECTIVE: To investigate the expression and clinical significanceof hypoxia inducible factor-1a(HIF-1?) and particularly interestingnew cysteine-histidine rich protein(PINCH) in esophageal squamous cellcarcinoma.METHODS: The expression of HIF-1? and PINCH in specimen of tumor and lymph nodes tissues was detected by immunohistochemical methodin61 patients without chemotherapy. The expression levels of HIF-1? and PINCH were measured by Western Blot in specimen of lymph nodes and different differentiation tumor.15 samples of adjacent normal esophageal mucosa were used as control.The relationship between the expression and clinical significance of HIF-1? and PINCHwas analyzed.RESULTS: HIF-1?was mainly overexpressed in tumor cell cytoplasm and nucleus,PINCH mainly expressed in tumor stroma,hardly expression in tumor cell, and the expression levelswereboth significantly higher than that in normal esophageal mucous tissues. Both HIF-1? and PINCH were highly expressed in metastatic lymph nodes.There were expression differences of HIF-1? and PINCH in different differentiationof esophageal squamous cell carcinoma.The lower the degree of differentiation, the higherexpression level and expression rate of HIF-1?and PINCH,also they were bothsignificantly higherin cases with lymph node metastasis than those without metastasis group. The positivecorrelation was found between HIF-1? and PINCHexpression in tumor and lymph node tissue.The above comparison of the data were statistically significant(p <0.05).CONCLUSION:1. HIF-1? and PINCH over-expression was found in the lymph nodes and tumor tissue of patients with esophageal squamous cell carcinoma, bothof them may play an important role in tumorigenesis, development and metastasis. 2. The expression level and rate of HIF-1? and PINCH in tumor cells wereraisedwith the reducing degree of tumor differentiation, and they were bothoverexpression in tumortissue with lymph node metastasis,indicating that they may be correlated with tumorigenesis, developmentand metastasis.3.The expression of HIF-1? was positively correlated with PINCH,they maybe target geneeach other and mutual regulation in tumorigenesis and progression.
