Genistein is an important non-nutritious element of soybean isoflavones. Many studies confirm that it can inhibit many kinds of human tumor such as gastric cancer, breast cancer and colon cancer both in vitro and vivo. RNA interference is a new technique that proved to be effective for suppressing gene expression, and has been applied for anti-cancer studies.To research the effect of combination of genistein with RNA interference on the proliferation and apoptosis of DU-145 cells, the effect of genistein on the proliferation of DU-145 cells was detected by MTT, cytometry and the apoptosis cells were observated from fluorescence microscope; A survivin siRNA plasmid was constructed, the expression change was examined by reverse transcription polymerase chain reaction (RT-PCR) after the plasmid had been transfected into DU-145 cells;The effect of combination of genistein with RNA interference on the proliferation and apoptosis of DU-145 cells was detected by MTT method and fluorescein activated cell sorter respectively. The results were as following.As the concentration of genistein and/or the reaction time increasing, the relative activity of both DU-145 and Hela cells decreased significantly. Genistein of the same concentration performed higher inhibition effect on Hela cells than on DU-145 cells; The relative activity of recombinant plamid-transfected group was lower than that of genistein-treated group, and the cells of combination group had the lowest relative activity. Recombinant plamid-transfected group induced more cell apoptosis than genistein-treated group, and the effct of apoptpsis induction of combination group was significantly higher than that of either group. When combination treated, the effect of apoptosis induction was enhanced as the concentration of genistein increased.In conclusion, genistein can inhibit the proliferation of DU-145 cells induce apoptosis in dose and time dependence. The combination of genistein with RNAi can significantly inhibit proliferation and induce apoptosis in DU-145 cells.
