In this dissertation the preparation method, the biological effect of antifatigue and hypoglycemic of Lycium ruthenicum fruit polysaccharide were studied. The results were summarized as follows:1. The optimum conditions for the removal of saccharide by 80% ethanol and the extraction of Lycium ruthenicum fruit polysaccharide by hot water were established. The effects of extraction temperature, ratio of solid and liquid and extraction time of saccharide were investigated. The optimum conditions were: extraction temperature 70℃, ratio of solid and liquid 1:8 and extraction time 40min; The effects of extraction temperature, extraction time and extraction times of Lycium ruthenicum fruit polysaccharide were investigated. The optimum conditions were: extraction temperature 90℃, extraction time 60min and extraction times 3 times. The yield of Lycium ruthenicum fruit polysaccharide reached (3.67±0.31)%, the content of polysaccharide was 46.62% under the conditions established.2. The antifatigue function of Lycium ruthenicum fruit polysaccharide approved. Feeding the mice with Lycium ruthenicum fruit polysaccharide of low, middle and high dosage, then respecting comparing with control group, it was found that Lycium ruthenicum fruit polysaccharide could obviously prolong the swimming time of loaded mice, enhance the SOD activities of mice blood and liver, decrease the MDA content of mice blood and liver, increase the content of liver glycogen and muscle glycogen, and decrease the blood urea nitrogen and blood lactic acid in mice after swimming. The results indicated that Lycium ruthenicum fruit polysaccharide had the antifatigue function.3. The hypoglycemic function of Lycium ruthenicum fruit polysaccharide approved. Feeding the mice with Lycium ruthenicum fruit polysaccharide of low, middle and high dosage and respecting injecting with alloxan, the blood glucose of polysaccharide group was obviousely lower than diabetic model group; feeding the diabetic mice (blood glucose content beyond 11.1 mmol/L) with Lycium ruthenicum fruit polysaccharide of low, middle and high dosage, then comparing with diabetic model group and metformin hydrochloride group, it was found that Lycium ruthenicum fruit polysaccharide could significantly reduce the level of blood glucose, enhance the activities of SOD in diabetic mice blood and liver,decrease the content of MDA in diabetic mice blood and liver,promote the conversion of blood glucose into liver glycogen. The results indicated that Lycium ruthenicum fruit polysaccharide had the effect of prevention and treatment on hyperglycemia, and effectively enhance the ability of anti-oxidation in the diabetic mice induced by alloxan. In addition, Lycium ruthenicum fruit polysaccharide could release hyperglycemia symptom that diabetic mice drunk and ate more and more but its weight was less and less.
