Objective: Study whether Lycium ruthenicum Murr.in Qaidam basin reduce photo-damage of ultraviolet(UVB)on HaCaT cells and explore its possible mechanism.Method: The polysaccharide extracted by using hot water extraction followed by ethanol precipitatio assisted with ultrasonic.HaCaT cells were randomly divided into a control group,UVB radiation group,experimental group.Lycium ruthenicum Murr.polysaccharide was added into the culture medium in the experimental group before 6 h of UVB irradiation.After 12 h,cells were observed by microscope.Collected cells proliferation was detected by MTS.Cells apoptosis ratio was detected by Flow Cytometer.Malondialdehyde(MDA)content test kit,superoxide dismutase(SOD)activity test kit,glutathione(GSH-Px)content test kit and catalase(CAT)activity test kit were used to detect SOD activity,GSH content,MDA content,and CAT activity.IL-1 content,TNF-? content was detected by Elasi.The Protein expression level of P16 was detected by Western Blotting.Results: Compared with the control group,the HaCaT cells were blurred and the death and floating were observed.The apoptotic rate increased,the cell proliferation rate decreased,SOD activity,GSH-PX content,CAT activity decreased,MDA content increased significantly,IL-1 and TNF-? levels in cell culture supernates and TNF-? in cell lysates were significantly increased,The Protein expression level of P16 increased in UVB group,and there were significant difference(P<0.05).In experimental group and UVB group,the morphology of cells was restored;the number of dead cells was reduced,the cell proliferation rate increased,the apoptotic rate decreased,MDA content were decreased,SOD activity,GSH content,CAT activity were increased,IL-1 content in cell lysates was increased,TNF-? content in cell lysates and cell culture supernates were decreased,the Protein expression level of P16 decreased,and had statistically difference(P <0.05).Conclusion: Lycium Ruthenicum Murr.polysaccharides in Qaidam basin is effective in protecting skin against UVB-induced skin damage by decreasing ROS overproduction subsequent excessive inflammation reactions and the the Protein expression level of P16.
