Objective: To determine of the contents of procyanidin B2 and total cyanidin in Lycium ruthenicum Murr., and to provide the quantity gist for Lycium ruthenicum Murr. To investigate the effect of the Lycium ruthenicum Murr Water extract on hypoxia at normal pressure tolerance, and to lay theoretical foundation of Lycium ruthenicum Murr against radiation. Finally, to study of Protective Effect of Lycium ruthenicum Murr. Against Radiation Injury in Mice and mechanism study.Methods: The total cyanidin was determined by UV-Vis spectrophotometry. The procyanidin B2 was determined by RP-HPLC on a Diamonsil C18 column(250 mm × 4.6 mm, 5 ?m) with the mobile phase of acetonitrile and 2% acetate solution under the gradient elution condition. The wavelength was 280 nm, and the flow rate was 1 ml/min. In Normal-pressure anti-anoxia experiment, SPF mice were divided into control,positive, Lycium ruthenicum Murr Water extract high,middle and low does groups(LH,LM,LL).Hypoxia at normal pressure experiments after mice were given intragastric administration Lycium ruthenicum Murr Water extract for 14 days.The Survival time,the hemogramand RBC,the NO content,the the activity of antioxidtion enzyme,the brain water content were observed. In against radiation experiment, SPF mice were random Ly divided into control group, model group,positive drugs group and L.ruthenicum high does,L.ruthenicum middle does,L.ruthenicum low does treatment group that experimented 3 days after irradiation,in the same way,experimented 3 days and 7 days after irradiation respectively. L.ruthenicum extract were administered orally to the mice in the three treatment groups and normal saline were administered orally to the mice in the control group, model group for 14 days. Positive group were treated with radioprotective agent amifostine(WR-2721) at 30 min before irradiation. Except the control group, other groups mice received quantity of 5 Gy X-radiation whole body evenly with one time. Hemogram,organ index,DNA,Caspase-3,Caspase-6,p53 contents were observed at 3d,7d,14 d after irradiation.Results: The procyanidin B2 showed good linearity from 1 to 50 ?g/ml. The content of total cyanidin and procyanidin B2 was 0.639 g/100 g and 0.141 g/100 g, respectively.Lycium ruthenicum Murr Water extract could significantly increase the Survival time in mice, as well as reduce the brain water content, while the HGB, RBC, CAT, GSH-PX, T-AOC have no significantly statistical difference.L.ruthenicum could significantly increase the total red blood cell count,hemoglobin count and DNA contents(P<0.05). The index of spleen had significantly recover 3days and 14 days after irradiation(P<0.05). L.ruthenicum low does group could significantly reduce Caspase-3 and Caspase-6 of serum in mice after 3days,7days,4ays after irradiation and L.ruthenicum middle does group reduced Caspase-6 of serum in mice 7days after irradiation. L.ruthenicum could decrease the expression of p53, protect the small intestinal epithelial cells, reduce the radiation injury of small intestinal. The effect of L.ruthenicum on antioxidant enzyme is different at 3d, 7d, 14 d after radiation.Conclusion: L.ruthenicum contain abundant procyanidin B2 and total cyanidin. L.ruthenicum could reduce the hypoxic injury, decrease Apoptosis of radiation injured mice.L.ruthenicum had protective effects on mice radiation damage.
