Objective TO investigate the expression of osteopontin (OPN) and MMP-2 inepithelial ovarian tumor.Methods Immunohistochemical staining S-P methods was used to detect the expression of OPN and MMP-2 in 10 cases of normal ovarian tissue, 10 cases of benign ovarian tumor , 7 cases bordline ovarian tumor and 50 cases epithelial ovarian carcinoma.Results Both OPN and MMP-2 were negative expression in normal ovarian tissue. The positive expression of OPN and MMP-2 in 10 cases of benign ovarian tumor were 0 case,2 case respectively; in 7 cases of bordline ovarian tumor were 6 cases, 5 cases respectively; in 50 cases epithelial ovarian carcinoma were 45 cases,42 cases respectively. The positive rate both OPN and MMP-2 were significantly higher than normal ovarian tissue,benign ovarian tumor and bordline ovarian tumor(p<0.05) . In 50 cases epithelial ovarian carcinoma, the positive expression of OPN and MMP-2 were 10/12,10/12 in G1 16/18, 14/18 in G2; 19/20,18/20 in G3.with the higher grade, the positive expressions of OPN were higher in every grade ,there was significant difference (P<0.05 ) . The expression of OPN was related with histological grade(rS=0.349,P=0 006<0.05). The expression of MMP-2 did not appear significant difference in every grade (P>0.05). The positive expression of OPN and MMP-2 were also found 25/27, 24/27 in serous; 16/18, 15/18 in mucinous, 4/5, 3/5 in endometrioid. There were not significant difference in different histological type(p>0.05 ) . The positive expression of OPN and MMP-2 were found 30/34,27/34 in clinical I—II; 15/16,15/16 in clinical III—IV.With the higher cinical stage , the positive expression of OPN and MMP-2 were higer in every stage,there were significant difference (P<0.05) . The expression of OPN and MMP-2 were related with clinical stage(rS=0.401, rS=0.383, P<0.05). The positive expression of OPN and MMP-2 were found 28/32, 26/32 in negative lymde matasis; 17/18,16/18 in lymde matasis. In each group, there were significant difference (P<0.05 ) . The expression of OPN and MMP-2 were related with lymde matasis (rS=0.401, 0.317, P<0.05). The expression of OPN and MMP-2 were correlated with ovarian carcinoma tumorigenesis (r=0.650 p<0.05) .Conclusion OPN may be a important marker for epithelial ovarian carcinoma.The expression of OPN was related with clinical stage, histological grade and lymde matasis (p<0.05) .There were not correlation between OPN expression and histotype(p>0.05). The expression of MMP-2 was related with clinical stage, and lymde matasis (p<0.05) .There were not correlation between MMP-2 expression and histotype and histological grade (p>0.05 ). OPN and MMP-2 may act cooperatively in epithelial ovarian carcinoma. Through inhibiting the OPN- MMP2 transmission path, We maybe find a new way to treat ovarian carcinoma.
