BACKGROUND AND OBJECTIVE: Kai1/CD82 was recently detected as a tumor metastasis suppressor gene. It silencing contributes to progression and infiltration of some tumors. Our study was designed to investigate the expression of Kai1/CD82 in nephroblastoma, and explore its correlation to clinicopathologic characteristics and prognosis of patients with nephroblastoma.Method: The immunohistochemistry was used to detect the expression of Kai1/CD82 in 48 specimens of nephroblastoma with diffirent stages. Clinical data and follow-up data of the 48 patients were analyzed. we selected out 48 cases of nephroblastoma and detected the level of KAI1/CD82 by immunohistochemistry.. They were divided into two groups, which the first was nephroblastoma group without metastasis including 34 cases, the second was neuroblastoma with lymph node metastasis or distant place metastasis group including 14 cases. And we put 11normal renal issues as as control group including 11cases. Then we detect the expression level of CD82, the protein expression product of KAI1 gene, in each group by immunohistochemistry method.Result: Posive rate of Kai1/CD82 was significantly higher in control group and the first group than in the second group. Its expression was negatively correlated to clinical stage of nephroblastoma, the positive rates of the expression of nephroblastoma with and without metastasis are 55.88% and 35.71%, respectively. It is statistically signifinant between the two groups. The position rates of KAI1/CD82 inâ… ,â…¡,â…¢andâ…£stages are respectively 60.00%,56.25%,42.86% and 37.50%. There are no statistical significances between every two groups. The differences of the sets with and without chemical therapy in the two groups are not significant either.CONCLUSIONS: The change of Kai1/CD82 expression is an early event in tumorigenesis of nephroblastoma. Its down-regulation may be considered as a potential indicator to judge the differentiation and metastasis of nephroblastoma., which can serve as one of the combined indexes to clinical assessment of proress. the position rate of KAI1/CD82 in nephroblastoma is related with metastasis, while it has nothing with clinical stage and chemical therapy. After analyzing the experiment rusult we found that the expression level of KAI1/CD82 changes into double phase, or we can say it express lowly in benign ganglioneuroma, but high in non-metastatic neuroblastoma and lowly again in nephroblastoma with metastasis. So we think that the KAI1/CD82 high expression of nephroblastoma in the early age may be associate with the inhibition of tumor malignant process, once KAI1 gene is defect or its expression level decreases, metastasis of tumor may take place. Otherwise the fact that the expression of KAI1/CD82 is associated with the metastasis of tumor shows that KAI1/CD82 can be regarded as a index on judging the prognostic of nephroblastoma. It also can help to select clinical treatment plan.
