Kai1/cd82 Expression In Neuroblastoma In The Discussion On Significance And Mechanism Of Action

Objective: The mechanism of high metastasis rate of neuroblastoma especially metastasis to bone marrow is still unknown.We detected the expression level of CD82 which is a kind of protein expression product of the Non-metastatic gene named KAI1 in different neuroblastoma group, so as to investigate the possible mechanism about KAI1/CD82 in neuroblastoma metastasis and significance in clinical treatment.Method: We select 9 definite ganglioneuroma cases from pathology department in affiliated hospital of Qingdao medical college from 1995 till now and 40 cases neuroblastoma paraffin block samples with each stages.They were divided into three groups,which the first was ganglioneuroma group including 9 cases,the second was non-metastatic neuroblastoma group including 21 cases,the last was neuroblastoma with lymph node metastasis or distant place metastasis group including 19 cases .Then we detect the expression level of CD82,the protein expression product of KAI1 gene,in each group by immunohistochemistry method.Result:The positive expression of CD82 was different in each group,2 in ganglioneuroma group(masculine rate is 22.22%), 15 in non-metastatic neuroblastoma group(masculine rate is 71.43%),6 in neuroblastoma with lymph node metastasis or distant place metastasis group(masculine rate is 34.74%).The difference between the former two groups shows statistical significance (P (0.05) ,so as the difference between the second and the third group (P (0.05 ) .If these neuroblastoma samples were divided by their clinical stage,then the masculine rates would be 80%, 40%, 62%,35.71%.We couldn't find statistically significant between either two groups.The expression level of KAI1/CD82 in neuroblastoma is associated with metastasis,while it isn't correlated with chemotherapy and metastasis including metastasis on bone marrow.Result:After analyzing the experiment rusult we found that the expression level of KAI1/CD82 changes into double phase,or we can say it express lowly in benign ganglioneuroma, but high in non-metastatic neuroblastoma and lowly again inneuroblastoma with metastasis.So we think that the KAI1/CD82 high expression of neuroblastoma in the early age may be associate with the inhibition of tumor malignant process,once KAIl gene is defect or its expression level decreases,metastasis of tumor may take place. Otherwise the fact that the expression of KAI1/CD82 is associated with the metastasis of tumor shows that KAI1/CD82 can be regarded as a index on judging the prognostic ofneuroblastoma.lt also can help to select clinical treatment plan.
