Objective In this study, we investigate the association between the level of PAI-1in plasma with type 2 diabetes with early nephropathy,and the association between thePAI-1 gene 4G/5G polymorphisms with type 2 diabetes with early nephropathy onmolecular level,and to explore related virulence gene.Method The level of PAI-1 in plasma was assayed by ELISA, the PAI-1 promoter4G/5G polymorphism in the control group (39 subjects) and the study group (63 patients)were genetyped by allele specific polymerase chain reaction (PCR).Results①There were significant differences in PAI-1 genetypes frequency and4G/5G allele frequency between the Patients with type 2 diabetes with early nephropathywith the normal group (?~2=12.186, P<0.05;?~2=7.446, P<0.05)and between the DM groupwith the normal groups (?~2=24.127, P<0.05;?~2=7.312, P<0.05). There was no differencesin PAI-1 genetypes frequency and 4G/5G allele frequency between the Patients with type2 diabetes with early nephropathy with the DM group (?~2=2.810, P>0.05;?~2=1.189,P>0.05).Patients with type 2 diabetes with early nephropathy and the DM group had a highfrequency of 4G allele frequency than that in the normaI group (?~2=6.280,P<0.05).②Inthe control group,the female subgroup do not significantly different from the malesubgroup in the distribution of PAI-1 gene and its allele frequency (C=8, P>0.05;?~2=0.320,P>0.05).③In the DM group,the female subgroup do not significantly different from themale subgroup in the distribution of PAI-1 gene and its allele frequency (C=3,P>0.05;?~2=3.724, P>0.05).④In the type 2 diabetes with early nephropathy group,thefemale subgroup do not significantly different from the male subgroup in the distributionof PAI-1 gene and its allele frequency (C=3, P>0.05;?~2=0.002, P>0.05).⑤In the type 2diabetes with early nephropathy group,the PAI-1 genetype polymorphism was relatedwith the albumin excretion rate (F=4.922, P<0.05).⑥The presence of the 4G allele ofPAI-1 in the DM groups(n=63)was involved in the generation of diabetes at 2.073 ratescompared with that in the normal group (?~2=6.280, P<0.05).⑦The plasma PAI-1 level inthe type 2 diabetes with nephropathy group and in the DM group were higher than that in the normal group(F=312.6066, P<0.0001),the PAI-1 level in plasma was significantlyhigher in 4G allele homozygous than those in 4G/5G beterozygous and 5G homozygous(F=21.0190, P<0.0001).There was no differences of the PAI-1 level in plasma with4G/5G polymorphisms in the control group (F=2.5460, P>0.05).Conclusion Elevated plasma PAI-1 level is a risk factor for diabetic nephropathy,there was a significant relationship between the PAI-1 gene 4G/5G polymorphisms withthe level of PAI-1 in plasma,and the presence of the 4G allele of PAI-1 may involved inthe generation of diabetes, the 4G allele of PAI-1 is a risk factor for albuminuria. |
