Background:Acute Spinal Cord Injury(ASCI) is a kind of high deformity rate disease. It's also the familiar wound in the orthopedics realm. Its treatment's as a medical science hard nuthas been paid attentio to by the international community experts. Recent years, with the development of modern transportation and building industry, Its outbreak rate always high. So investigating a new method to cure the spinal cord injury suffererhave important medical treatment worth and social meaning. Ulinastatin Is a kind of broad-spectrum enzyme inhibitor. It has a function of repress to various biocatalyst and has already applied extensively in clinical. This topic studies the experimental research about the influence of using Ulinastaitin to adjust the micro environment in spine cord injury and the protective function on nerve cell. Through observing spinal cord dropsy, Inflammatory reaction, lack of blood and anoxia's etc,the changes of pathologic physiology, we investigate and confirm the utility effect on spine cord in using Ulinastaitin.Method:Eighty SD rats were randomly divided into four groups: acute spinal cord injury groups ( SCI groups, n=20);administration of Ulinastatin (U groups, n=20), use Ulinastatin after SCI at half an- and 4- and 8- and 24- and 48- hour.;administration of Methylprednisolone (M groups, n=20), use Ulinastatin after SCI at half an- and 4- and 8- and 24- and 48- hour; administration of Ulinastatin and Methylprednisolone(M+U groups, n=20), use Ulinastatin and Methylprednisolone after SCI at half an- and 4- and 8- and 24- and 48- hour;Allen's weight drop model of acute spinal cord injury was prepared. BBB scales and Molt's inclined plane scales, pathological changes of spinal cord and motor evoked potentials at 24- and 72-hour and 1- and 2- and 3-week after SCI were respectively studied.Results:Express as a result through research:in BBB scales and Molt's inclined plane scales, pathological changes of spinal cord and motor evoked potentials measurement,we easily found that 3 using medicine groups surpass the control group. Among them, the effectiveness on spinal cord injury rats in using Ulinastatin group is not as good as using Methylprednisolone group after injury in 24 hours, but after 72 hours, Ulinastatin group had the equivalency result with Methylprednisolone group. In 3 weeks, Ulinastatin group's result was slightly the good to Methylprednisolone group, but have no statistics difference. In each time segment Ulinastatin group's result was obvious good to control group, and it had patency statistics difference. Drug combination group had the best instauration result in each groups. Methylprednisolone group died 2 rats because of the alimentary tract hemorrhage. Control group died 2 rats because of the bloody urine. The rats in Ulinastatin were all safe in our experiment.Conclusion:The research result of our experiment hints that the mechanism of action and action time of Ulinastatin has certain differentiation with Methylprednisolone. The limited efficacy of action time of Methylprednisolone is quick, but But its valid time is short, and it has great adverse reaction. In initial stage, the effectiveness of Ulinastatin is not as strong as Methylprednisolon, but it has the features of longer action time and higher safty than Methylprednisolon, and because of synergistic action with Methylprednisolon Ulinastatin can produce huge effect on curing ASCI. Our experimentprovide a theories basis for clinical in curing acute Spinal Cord Injury.
