ObjectiveThe model of insulin resistance was made by high-fat diet. The level of insulin and hsCRP were detected and the expression of NF-κB and MCP-1 were observed in endothelial cells. To observe the change of MCP-1 and factors which caused the changing in high-fat diet rats, and to evaluate its action in arteriosclerosis accelerated by insulin resistance.Methods16 male Wistar rats were randomly assigned into 2 groups: normal diet group and high-fat diet group after 1-week-accommodation raising. Every rat was weight once a week. Each group was fed for 8 weeks. IR was verified by euglycemic-hyper-insulinemia clamp technique. Common carotid artery were taken for immunohistochemistry detection. Blood from left ventricle was taken for the detection of blood serum's factors. Level of insulin and hsCRP were determined and the expression of NF-κB and MCP-1 in artery endothelial cells was examined by immunohistochemistry. The differences of these factors between the two groups and the correlation between the factors linked to IR and atherosclerosis were respectively analyzed by statistics software SPSS 13.0.Results1. IR was verified in high-fat diet group by using euglycemic-hyper-insulinemia clamp technique, but not in normal diet group. In normal diet group, FBG was 5.01±0.20mmol/L, the level of insulin was 1.41±0.44mmol/L and GIR was 14.41±0.69 mg/kg.min. In high-fat diet group, FBG was 5.33±0.20mmol/L, the level of insulin was 2.23±0.85mmol/L and GIR was 9.40±1.29mg/kg.min. 2. In high-fat diet group, the level of hsCRP was markedly increased compared with normal diet group (P<0.05). The level of hsCRP in high-fat diet group was 2.14±0.39mmol/L, which was 1.63±0.23mmol/L in normal diet group.3. In high-fat diet group, the NF-κB and MCP-1 were significantly expressed in artery endothelial cells. The rate of coloring with NF-κB and MCP-1 were 0.44±0.05 and 0.90±0.07. But in normal diet group, there is little expression. The rate of coloring with NF-κB and MCP-1 were 0.01±0.02 and 0.11±0.24.4. The expression of MCP-1 was positive correlated with the level of insulin (r=0.503,P<0.05). And the level of hsCRP was also positive correlated with the expression of NF-κB (r=0.701,P<0.05). The relationship between MCP-1 and NF-κB was positive too (r=0.932,P<0.05).Conclusions1. The increase of insulin stimulates the MCP-1 in IR.2. In IR, inflammatory factors and oxidative stress indirectly made the expression of MCP-1 increased by NF-κB.
