Objective: To investigate the protective effect of vitamin E to retina in rabbit with experimental vitreous hemorrhage.Methods: 60 Newzealand white rabbits were divided into groups A, B and C, each group was 20 rabbits.Group A was sham operation group which intravitreal injected 0.2ml physiological saline; Group B was model control group and group C was vitamin E treatment group which intravitreal injected 0.2ml autologous blood. Groups A and B were subcutaneously injected 0.04ml/Kg (40mg/Kg) physiological saline once a day; Group C was subcutaneously injected Vitamin E 40mg/Kg once a day. Randomly killed four rabbits in each group on the 3rd,7th,14th,21st and 28th day. 2 eyes of each group were fixed by 4 percent of paraformaldehyde, made into pathological sections. Others' retina was separated quickly, made into 10 percent homogenate, to determine the changes of MDA, the activity of SOD and Na~+-K~+-ATPase.Results: 1. On the 14th day: It was found ganglion cells vacuolar degeneration, inner plexiform layer thickened locally in group B. No obvious degeneration was observed in group C. On the 21st day: It was found ganglion cells,inner and outer segments of photoceptor,inner and outer nuclear layers hydropic degeneration, the inner layer of retina became thick in group B. No obvious degeneration was observed in group C. On the 28th day: It was found ganglion cells hydropic degeneration and the inner layer thickened obviously. Red blood cells and its fragments moved into the retina, and the proliferation was observed on the retinal surface in group B. It was found ganglion cells and inner nuclear layer hydropic degeneration, the inner-layer thickened. Few red blood cells was found in the retina and no obvious proliferation was observed in group C. 2. On the 7th,14th,21st and 28th day, the MDA level in groups B and C significant higher than that of group A, and group C significant lower than that of group B.3. On the 7th,14th,21st and 28th day, the acitivty of SOD in group B significant lower than that of group A, group C significant higher than that of group B. 4. On the 14th,21st and 28th day, Na~+-K~+-ATPase activity in groups B and C significant lower than that of group A. On the 21st and 28th day, Na~+-K~+-ATPase activity in group C significant higher than that of group B.Conclusions: 1. Vitreous hemorrhage can cause retinal cells hydropic degeneration,epiretinal membrane formation and lipid peroxidation to result in retinal damage. 2. Vitamin E can relieve the hydropic degeneration of rabbits' retinal cells and the lipid peroxidation of retina, delay the process of proliferation in vitreous hemorrhage. Vitamin E has a certain protection effect to retina.
