| Objective1. To investigate the relationship between the expression of glucocorticoid receptor and the quantity of CD4~+CD25~+Treg in patients with depression, and to study the relationship between the receptor and Foxp3, which is the characteristic marker of CD4~+CD25~+Treg. Then the result can be used to explore the possible effect of immune disequilibrium, caused by glucocorticoid receptor and CD4~+CD25~+Treg.2. To explore changes of cytokine IL-10 TGF-βin depressed patients' serum and to study these cytokines' roles in the immune disequilibrium caused by glucocorticoid receptor and CD4~+CD25~+Treg.Method1. On the basis of CCMD3 and DSM-IV, patients with depression are picked up. SDS and Hamilton depression rating scale are used to evaluate the level of depression. There are 4 groups in the research: normal, mild (9 samples), moderate (12 samples), and severe (15 samples). There is no statistic difference among these groups.2.The expression of GRαGRβFoxp3mRNA are detected by RT-PCR withβ-actin as internal control. 3. CD4~+CD25~+Treg are detached from PBMCs by Immunomagnetic beads and are counted with the microscope.4. The concentrations of IL-10,TGF-βfrom samples' serum are detected by ELASA.5. The immunofluorescence technique and the laser scanning Confocal microscope are used to observe the co-expression of Foxp3 and GR directly.6. Validating the homoscedasticity before using analysis of variance. Correlation analysis and S-N-K comparison are also used to analyze results.Result1. When it is compared to normal one (20.0±2.6×10~7/L, 5.80±1.26 %) , CD4~+CD25~+Treg Treg's quantity (8.3±1.6×10~7/L) is less and its proportion in CD4~+T subset (3.01±0.69%) is lower in depression groups, especially severe one. And its quantity and proportion in CD4~+T subset decreases when the depression becomes serious.2. The expression of GRαmRNA Foxp3mRNA in depression patients is significantly lower than normal ones. However, almost every expression of GRβmRNA is negative in the normal, mild and moderate group, except one in each group. In severe group, there are 9 samples expressing positively.3. The concentrations of IL-10 TGF-βare decreased in depression groups, especially severe one.4. GR expresses in cytoplasm and nucleus, when Foxp3 expresses in nucleus. Compared with normal one, both GR and Foxp3 express less in CD4~+CD25~+Treg in severe group.5, After correlation analysis, there are correlativity in the data as follows: Foxp3 mRNA and GRamRNA; Foxp3 mRNA and the quantity of CD4~+CD25~+Treg; Foxp3 mRNA and the concentration of IL-10/TGF-β.Conclusion1. There is the immune activation in depression happens. The decreasing expression GR plays an important role in the immune disequilibrium.2. GR, existing on CD4~+CD25~+Treg, impacts the expression of Foxp3 which induce the dysfunction and decreasing of CD4~+CD25~+Treg, finally result in immune activation in depression.3. IL-10 and TGF-βhave significant effect on the neuro-endocrine-immune system which impacted by GR and CD4~+CD25~+Treg Treg in patients with depression. |
