Objective:After several years of study,we think that psoriatic bone marrow hematopoietic cells are abnormal.In order to study their function in psoriasis further,we are going to transplant psoriatic bone marrow to SCID mice.But this research involves many difficult problems of the present immunology and hematology which are not yet completely solved.How to promote the hematopoietic and immune reconstruction of heterogeneous bone marrow and the long-term survival of SCID mice are both difficult problems which must be solved.To construct an advantageous micro environment of the growth and differentiation of the human T cell in mouse, we must transplant thymus lobes to acceptor while carrying out xenogeneic bone marrow transplant.It is necessary to remove the original thymocytes from the thymus lobes to ensure the maturation of hematopoietic cells in the acceptor,since the original thymocytes would interfere with immune reconstruction.In addition,the immune rejection is the primary factor which affects the animals' living conditions after xenogeneic bone marrow transplant.We can learn the primary cause of death of the animals in the immune rejection by observing their living conditions.Carrying on processing to the animals in view of this reason can reduce the intensity of immune rejection.Thus the animals' life may be lengthened maximumly.Therefore this topic mainly studies the following two aspects:To explore the ideal method of removal of original thymocytes from the thymus lobes by comparing the action of different culture conditions on removing thymocytes from thymus lobes.To explore the direct cause of death of the SCID mice by observing their living conditions after transplanting psoriatic bone marrow to them.Methods:Aborted embryonic thymic lobes were cultured in RPMI-1640 medium supplemented with 1.35mmol/L 2'- deoxyguanosine on gas-liquid interface,using thymic lobes cultured in medium without 2'-deoxyguanosine as controls in parallel.The thymocytes in cultured lobes were observed by HE staining at different culture period respectively.Cultured thymic lobes and psoriatic bone marrow were both transplanted to SCID mice.The animals' living conditions were observed.Results:The original thymocytes reduced significantly after cultured in the medium containing 2'-deoxyguanosine and nearly vanished after 5 days,while there is almost no decrease for thymocytes count in the first 10 days of culture process for the controls.The thymocytes all obviously reduced both in the test group and the comparison group after being cultured for 20 days,and the matrix also appeared the varying degree necrosis.With the lengthening of culturing time,the necrosis degree also aggravated.Seven cases died in 7 day after transplantation,the cause of death were all intestines necrosis.Two cases gradually returned to normal after a transitory response and has survived so far(35 days after transplantation).One control mouse has no abnormal reaction and has survived by now.Conclusions:It is an ideal condition to remove original thymocytes in thymus lobes by culturing the lobes in the culture medium containing 2'- deoxyguanosine for 5 days.Intestines necrosis is the primary cause of death of the SCID mice after transplanting heterogeneous bone marrow.
