Objective:To study the expression leves of SCCA gene in 30 squamous carcinoma of the cervix tissues and 15 normal cervix tissues by real-time RT-PCR.To investigate whether this gene plays a role in the pathogenesis of squamous carcinoma of the cervix.The correlation of expression of SCCA gene with clinicopathological characteristics of squamous cervical carcinoma was also analyzed. Methods:From April 2006 to October 2006,thirty patients with squamous cervical cancer and fifteen patients with hysteromyoma was selected in ShanXin Cancer hospital.Quantitative real time RT-PCR according to an internal control GAPDH was used to detect the expression of SCCA mRNA in 30 squamous carcinoma of the cervix tissues and 15 normal cervix tissues.Results:The expression level of SCCA2 mRNA in 30 squamous carcinoma of the cervix tissues was higher than in 15 normal cervix tissues(P<0.001),while those of SCCA1 mRNA did not significantly differ between normal and malignant tissues(P>0.05).The expression of SCCA2 mRNA was higher than SCCA1 mRNA in squamous carcinoma of the cervix tissues.The expression of SCCA2mRNA was relevant to FIGO stages,there was a tendency for them to increase with the stage increasing(P<0.001).Moreover,the overexpression of SCCA2mRNA was significantly correlated with lymph node metastases(P<0.05).The expression of SCCA2mRNA was no correlation with age and histological grade(P>0.05).However the expression of SCCA1mRNA was no correlation with age,FIGO stages,lymph node metastases and histological grade(P>0.05).Conclusion:The up-regulation of SCCA2 mRNA may be closely associated with tumor FIGO stages and lymph node metastases.Otherwise there was no relationship between the expression of SCCA1mRNA and the character of clinical pathology.Quantitation Real-Time Reverse Transcription-PCR with TaqMan probe assay expression of SCCA mRNA in squamous cervical carcinoma could be useful for individual diagnosis and prognosis observation.These data indicate that specific quantitation of the expression level of SCCA2 mRNA may be useful for the diagnosis and management of patients with squamous cervical carcinoma.If this finding can be confirmed in a larger study the SCCA2 mRNA in primary tumors could be useful for individual selection of treatment strategy for patients with squamous cervical carcinoma.
