Object:Elucidating the qualitative and relative quantification expression of Plkl in three groups:normal cervical tissues, cervical intraepithelial neoplasia(CIN) and cervical neoplasms,and on that condition,probing the relationship of Plkl with certain clinical and pathological features among cervical carcinoma tissues.Thus,the critically important mechanism role of Plkl may be attracted,and the biomarker-mediated entry from CIN to cervical cancer gained a strong evidence. At the same time,an interesting finding that its structure and function implications documented that was a promising target-specific therapeutic molecular.All of these hope it can become an effective predictor of diagnostic markers and detection.Methods:By means of Real-time fluorescence quantitative PCR and immunohistochemical staining SP technique tools utilizing in the report,this trial had two large steps.Small parts of one steps included extracting RNA,reverse transcription cDNA and Quantitative fluorescence PCR,when the standard procedure finished,levels of Plk1mRNA in collective ten cases of normal cervical tissues,twenty-six cases of collective cervical squamous cancers, and twelve cases of CIN collective groups was darkly putted out.Remaining collective tissues were Paraffin-embedded and made slices in Immunohistochemistry.Finally,the positioning and qualitative expression of Plkl protein was exploited by Immunohistochemistry. Data showed in different cervical tissues and relationship among age, clinical stage and the differentiation in cervical cancer must be correctly evaluated through statistical software.Results:It was interesting that the expression of P1k1mRNA and P1k1 protein was consistently ascending in carcinomas and CIN comparing normal cervical tissues,while CIN drop in cancers.Results of whole P1k1 data was importantly Statistical (P<0.05).Clinical age,pathology differentiation and staging were not all related to Plkl expressed in cervical tumors.1. Calculated levels of P1k1mRNA in collective cervical tissues by RT-PCR:Based on the slowly changes from normal to abnormal cervical tissues, the amounts of P1k1mRNA levels also changed in experimental groups. The cancers group was elevated than other two groups(CIN groups,normal groups) and gained exciting Statistical comparisons significance respectively(P<0.05). There has a rising trend compare CIN group to normal group and this difference stand by Statistical significant(P<0.05).2. Results of Plkl protein expressing monitored in different cervical tissues by SP:It was gradually coming up that plkl protein expression from normal cervical tissue to malignant cervical tissue. Samples such as normal cervical tissue,precancerous lesions and cervical squamous cell carcinoma,respectively,Plkl positive expression rates were 0%,50%, 88.5%. Obviously, there existed difference in three group and the up-regulated sequential difference was Statistical significant (P<0.05).3. The relationship between clinical pathological features and cervical neoplasms on two technique data:There was association of P1k1mRNA or Plkl protein expressed in the varying degrees of differentiation (P<0.05). High expression of PLK1 in tumor cells related to the level of differentiation,while not others.PLK1 differently expressed in the abnormal cervical tissues relative to control group may panel with plkl trial in the progressively development of cervical neoplasms and changes of pathological.Outcome ultimatly showed that it can be used as an valid predictive factor of early cervical cancer emerging,and may assist in the diagnosis and prognosis and the transferring of cervical cancer.It may be taken as an indicator event and indicated some clinical significance.
