Objective:Ischemic Acute Cerebral Infarction is a common disease.The high death rate and disabilitity is becoming a serious medical problem,how to reduce the fatality,promote the recovery of neurofunction,improve the life quality of patient are becoming a hot spot for neurologist.To provide proofs for the possibility of rhG-CSF treatment in ischemic stroke,we examined the effects of rhG-CSF on the functional outcome,cell recovery and proliferation after focal ischemia in rat brain.Method:A total of 108 androus Sprague-Dewley(SD)rats were divided into therapy group, control groupand and sham operation group randomly.Every groups were divided into 24 hours, 48hours and 1 week group.Following Koizumi's method,we subjected middle cerebral artery occlusion(MCAO)by processed fishing line.Therapy group and sham operation group were injected rhG-CSF in subcutis,then sacrificed at the time point including 24h,48h and lw after behavior tests.Flow cytometry(FCM)was used to detect the apoptotic.Hematoxylin and eosin (HE)stain and immunohistochemical techniques were exerted to inspect the morphous of neurocyte,the expression of CD34+ cell and neurological stem protein(Nestin)in infarct and peri-infarct area of the brain.We recorded all the experimental result for statistical analysis.Results:l The death rate of the rat was lower in G-CSF therapy group than control group (P<0.05).The infarction volume of the rat brain was smaller in G- CSF therapy group than control group in 24h,48h and lw later(P<0.05)and the pathological damage is slighter.There was no infarction area in sham operation group.2 Many new vessels were appeared in infarction area of the therapy group.Less apoptotic cell was found in rhG-CSF-treated animals compared with control animals(P<0.05).CD34+ cells were detected in infarction area of therapy group with nestin-cells lw later.Little new vessels,no CD34+ cells and nestin-cells were found in control group.Conclusions:Bone marrow stem cell was mobilized to infarct area and transformed into neurocyte and new vessels,Low mortality,diminished infarct area,reduced apoptotic cell were found in animals treated with rhG-CSF,suggesting rhG-CSF prompted neurofunction recovery.
