Objective:To observe the expression of phosphorylation IKK and phosphorylation IκBαin the great vessel of Diabetic rats and explore the relationship of IKK,IκBαand the great vessel lesion in diabetic,to provide the theoretical foundation for early diagnose and therapy of the macro-vascular complications in diabetic.Methods:48 Wistar male rats were divided randomly into two groups.There were 24 in normal control group(group A)and 24 in experimental group(group B).The subjects in group B were injected intraperitoneally with dose streptozotocin(STZ,50mg/kg)while the ones in group A were injected intraperitoneally with the similar dose of citrate buffer.We randomly killed eight rats of every group respectively at Six weeks,ten weeks and forteen weeks.The weight of every subject was measured.The blood samples were obtained to measure the levels of fat(TC,TG,LDL-C,HDL-C),FINS and C-peptide.The morphology changes of the carotid artery were examined by light microscope and transmission electron microscope,the carotid artery intima-media thickness(IMT)were measured,The expression of phosphorylation IKK and phosphorylation IκBαwere determined by Immunohistochemisty.Results:1.After 72 hours injected intraperitoneally with STZ,The fast blood glucose(FBG)of rats in group B was higher than 16.7mmol/L.There was a significant difference between group A and group B(P<0.01);and the level of of FINS and C-peptide of rats in group B was lower significantly than group A.2.After admimistrated with STZ ten weeks later,the carotid artery intima-media thickness(IMT)of group B was higher than that of group A(P<0.01),fourteen weeks later,IMT was higher than that of ten weeks;The levels of TC,TG,LDL-C and HDL-C of group B were different significantly than group A3.The expression of phosphorylation IKK and phosphorylation IκBαof group B were much higher than group A(P<0.01).the result of linear Correlations analyse showed:in group B the expression of phosphorylation IKK and phosphorylation IκBαwere positive correlation with IMT and the levels of TC,TG and LDL-C,and in group B the expression of phosphorylation IKK was positive correlation with the expression of phosphorylation IκBα.,but the expression of phosphorylation IKK and phosphorylation IκBαwere negative correlation with the levels of HDL-C.4.Morphology observation:light microscope and transmission electron microscope(TE) examination revealed that the damage of carotid artery appear at the stage of six-week in group B,more and more severe with the progress of diabetes mellitus.There was no morphologic change of carotid artery in group A.Conclusion:1.During the progress of great vessel lesion of diabetes mellitus,The level of phosphorylation IKK and phosphorylation IκBαin carotid artery was rising,and the results revealed that phosphorylation IKK and phosphorylation IκBαare the danger factor of great vessel lesion of diabetes mellitus.2.In carotid artery of the diabetes mellitus rat model,with the development of diabetes mellitus,The level of expression of phosphorylation IKK was rising,the result revealed that the expression of phosphorylation IKK was positive correlation with the expression of phosphorylation IκBα.and the results revealed that phosphorylation IKK is the key factor of great vessel lesion of diabetes mellitus.3.the expression of phosphorylation IKK and phosphorylation IκBαwere positive correlation with the levels of TC,TG,LDL-C.but the expression of phosphorylation IKK and IκBαwere negative correlation with the levels of HDL-C.the results revealed that the fat metabolism mess caused by hyperglycemia may induce the activation of IKK and IκBα.
