Objective To evalute the expression of MnSOD and E-cad in the Nasopharyngeal Carcinoma(NPC) inorder to investigate their roles in tumor's biological behavior and the prognosis of NPC and the correlation of them.Method Collected 60 NPC patients who were pathologically confirmed as poorly differentiated squamous cell carcinoma and had a paraffin block,all of them were radiotherapied with 4MV X-ray and 60Co r-ray. Therapeutic dose of the nasopharynx was DT66~72Gy/7~8W. Cases without lymphatic metastasis were given DT50~54Gy/5~6W to cervix. Cases with lymphatic metastasis were given DT66~72Gy/7~8W on the regional lymph nodes. The expression of E-cadherin and MnSOD was detected by S-P method of immunohistochemistry. SPSS 11.5 was applied to analyze the database.Results 1 The strong positive expression frequency of MnSOD in the NPC tissue was 46.7%. While E-Cad's was 48.3% ,and its strong positive expression in lymph-metastasis group was 41.1%,in nonlymph-metastasis group was 77.8%. 2 The positive expression rates of MnSOD was positively correlated to T stages(r=0.516,P=0.000),indicated the expression of MnSOD increase as tumor developed. The expression of E-Cad showed no positive correlation with T stages(P= -0.189,r=0.147),it suggested that E-Cad had no obvious effect on tumor's regional infiltration.3 MnSOD was significantly associated with N stages . But no obvious relation was found between MnSOD and the region of lymphatic metastasis. E-Cad was negative correlated to N stages(r= -0.386,P=0.002), and different levels were statistical significance(P<0.05). And E-Cad expression was closely relevant with the lymphatic metastasis rate,the two factors showed negative correlation(r=-0.469 , P=0.000).But the expression of it had no obvious effect on lymph node volume.4 Statistical analysis showed that the later their clinical stages were, the higher level the MnSOD was(P<0.05),and the MnSOD was positively correlated with clinical stages(r=0.303,P=0.009).While the E-Cad were on the opposite,Clinical stages were negative correlated to the expression level of it (r=-0.351,P=0.006).5 MnSOD highly expression resulted in the dose for complete reduction of nasopharyngeal primary tumor increased and the complete disappearance rate of lymph node decreased (P<0.05). The expression of E-Cad show no correlation with radiosentivity. 6 The survival rate was declined obviously along with enhancement of MnSOD's expression.While the group with high expression of E-Cad had a high survival rate, showing a good prognosis. Statistical analysed showed that all levels of expression had obvious significants.7 Cox harzed model analysis indicated that clinical stages and the high expression level of MnSOD were the risk factor affecting 5-year survival rate and the high expression level of E-Cad was the protective factor affecting 5-year survial rate.But no obvious correlation was found between them.Conclusion: 1 High expression of MnSOD might has correlation with NPC's regional invasion and lymph node develop. 2 High levels of MnSOD suppressed radiosensitivity. 3 Enhancement of E-Cad'expression supressed lymphatic me- tastasis but had no relationship with tumor's local infiltration. 4 The expression of MnSOD and E-Cad might play important role in terms of survival rate. 5 No significant correlation was found between the expression of MnSOD and E-Cad.
