When the lens is injuried, the aqueous will come into lens,the lens capsule or the cortex was broken .then the epithelial are edama. on the other hand, the lens will become obsque because lens fiber was degenerated or scar keloid formed. Finally the traumatic cataract (TC) happended. According to the documents that have been punished, the incidence rate of TA is more than 36%-52% during the penetrating eye traumas. Especially at the penetrating cataract, it will result in deformation of crystalline fiber and will destroy junction of cell-cell, these will aggravate the injure of epithelial cells, result in the metabolic disorder of epithelial cells, precipitate apoptosis.The lens epithelium cell is the most athletic part of metabolism in lens, it possess the function of crystal growth, differentiation and renovation. Therefore, any damage to endothelial cells will effect the normal physiologic function of lens, to cause the water retention in lens, finally induced cataract. In the process of cataract, the epithelial cells' hypofunction and the numbers to decrease, this is the result of apoptosis. The extracapsular cataract extraction and intraocular lens implantation after traumatic cataract can make patients to see the things again, but after operation, the epithelial cells will accruement and migrate, to result in opacitas of postcapsule. Seriously cause displacement of lens or grow blind again. So to study and research the effect of cortex opacitas to apoptosis of epithelium lentis cells after traumatic cataract will have a energetic effect to prevention and cure the PCO.This experimentation through establish the model of culture in vitro and the animal model to search for the relationship of cortex opacitas and apoptosis, to supply another study road of prevention and cure the PCO.Materials and methods1 To establish the traumatic cataract model of oculus bovinus that culture in vitro and divide into groups, put the oculus bovinus into procapsular group, postcapsular group, the aequator capsular group and negative control group, on the lens capsule, use pinhead perpendicular to cut 3-4 pieces wound, which diameter is l-2mm. then to revolve the pinhead, agita the cortex under capsular. take them into bioclean glasswares which have DMEM culture fluid . after cultivate 1,3,6,9,12 days , take out of the lens to observe and take the procapsular to stretched preparation. March giemsa dyeing, and treat it with tunel dyeing, and then calculate the rate of apoptosis.2 Establish the traumatic cataract model of hare's eye and divide into groupsTwenty-five healthy albino rabbits weighting 2-2.5kg were randomly dividedeinto 5 groups. One is being control group, the other groups were experimental groups. Established the mono-eye traumatic cataract models in experimental groups, after operation,use slit lamp microscope to observe the lens every day. at 12h,1d,2d,3d,5d,9d, take the procapsular to stretched preparation. March giemsa dyeing, and treat it with tunel dyeing, and then calculate the rate of apoptosis.Results1 The observation of bovine lens opacification at different time: 12h after injury appeared hazy opacity limitations; 3 days after injury, damage parts of the lens opacification extended color gray; 6 days after the injury, all the injuries were complete lens opacity, turbidity deepening; 9 days after the injury. The injury was obvious lens was milky white opacity. in the control group.the lens were transparent no turbidity there.2 Slit lamp microscope observed rabbit traumatic cataract model of Experimental eye injury appeared hazy opacity limitations; within 24 hours after injury in all experimental eye presence more visible reaction inflammation, the experimental eye lens opacification broadened significantly, maitain light white hazy. Injury after the 4th day before the room floc disappeared. All experimental eye with varying degrees of lens opacification, two of the limitations of turbidity, and the rest of the complete turbidity. Turbidity deepened, both white floc stale. 7 days after injury, the development of the more obvious cataract turbidity was massive white milk complete opacity, Since then showed progressive lens opacification. Control eyes, transparent lens, has no lens opacification.3 Giemsa staining bovine and rabbit lens epithelial cell morphology with time extension bovine and rabbit lens epithelial cells have shown for the former capsule under the epithelial cells decrease smaller cells, cell gap widened, defined border, karyopyknosis gradually increased.4 TUNEL staining and lens epithelial cell apoptosis rate 1 .Tunel staining, rabbit and bovine lens epithelial cells with the development of lens opacity, brown apoptosis cells gradually increased, while the control group showed no significant change5 At different times of the two groups of rabbits apoptosis rate data matching t test, t = 2.32 pm 1d, P<0.05;2d when t=2.48, P<0.05;3d when t=2.82, P <0.05; in different time bovine lens epithelial cell apoptosis four group rate for the single-factor analysis of variance, day 3 : F=6.36, P <0.05, Group 4 shows the difference was statistically significant; No. 6d : F=7.21. P <0.05, Group 4 shows the difference was statistically significant; No. 9d pm : F=9.32, P <0.05, Note 4 The difference was statistically significant; 12 days : The F=5.21, P <0.05, Note 4 The difference was statistically significant; 20th day : The F=7.24, P <0.05, Note 4 group with statistical significance. 3. Negative control group showed no change.Conclusions1 This study firstly establish the model of traumatic cataract of bovine lens in vitro.2 This study showed the cortex of lens opacitas improved lens epithelial cell apoptosis in traumatic cataract. The lens epithelial cell apoptosis arised with the increasing of the cortex of lens opacitas and the lapse of time.
