ObjectiveWe applied immunohistochemical technique to examine the expression of HIF-la andCD34 in nasopharyngeal carcinoma and nasopharyngitis tissues,evaluated the role ofHIF-la and MVD in nasopharyngeal carcinoma, probed the relationship betweenHIF-la and MVD.MethodsThe expression of HIF-la was evaluated in 58 cases with nasopharyngeal carcinomaand 20 samples of nasopharyngitis tissues by immunohistochemical staining SABCmethods and SP methods. The microvessel density(MVD) in tumor was counted byimmunostaining with CD34 and analyzed with clinicopathologic parameters.ResultsThe positive rate of HIF-la was 79.31% and 10% respectively in nasopharyngealcarcinoma and nasopharyngitis tissues, there was significant difference betweenthem(P<0.01). The microvessel density was 39.74±7.41 and 18.43±3.24 respectivelyin nasopharyngeal carcinoma and nasopharyngitis tissues, there was significantdifference between them(P<0.01).The expression of HIF-la and the microvesseldensity(MVD) were significantly higher in nasopharyngeal carcinoma and in lymphnode mesastasis group than in no-metastasis group.In different clinical stage, Theexpression of HIF-la(p<0.05)and MVD(P<0.01) was different.The expression ofHIF-la was positively correlated with MVD(P<0.01).ConclusionsThere was less expression of HIF-la in nasopharyngitis tissues.In nasopharyngeal carcinoma, HIF-1a expression was correlated with the tumor staging and lymphnodemetastasis. It accounts for that HIF-1a attach itself to the emergence and infiltrationand transfer of NPC. MVD was bigger in NPC than that in nasopharyngitis tissues.MVD was correlated with the tumor staging and lymphnode metastasis. These areexplained that MVD is get up vital function in the infiltration and transfer of NPC.MVD of HIF-1a positive group overtopped MVD of HIF-1a negative group in NPC.There was close correlation between the expression of HIF-1a and MVD. Thesemanifests that the expression of HIF-1a in NPC is close correlated to neoplasmangiogenesis and that HIF-1a play a vital role in the progression of NPC. HIF-1a islikely to a potential target of anticarcer therapy.
