Lycoris radiate (L.Herit) Herb is a kind of plant that distributes extensively in China, which is not only worth watching, but also can be used as pesticides and chemical raw materials. What's more important is its great medicinal value. Medical research has shown that galanthamine in its bulbs is a specific, competitive and reversible inhibitor of acetylcholinesterase, therefore galanthamine can serve as the treatment of Alzheimer's disease, and the effects to mild cognitive impairment were significantly. In addition, pseudolycorine, lycorine, polysaccharide and LRA have been respectively conformed as anti-tumor, antihypertensive, hypoglycemia and antiviral factors. Therefore, the comprehensive development and utilization of Lycoris radiate Herb. is imperative.The wild Lycoris radiate Herb. covers large areas in the south of our province. The local residents and the government has not truly understood its medicinal value, therefore, it has not been exploited. In this paper, while specifically studying the extraction, isolation and detection method of galanthamine by using the wild Lycoris radiate Herb. as raw material, we extracted pseudolycorine and polysaccharide as byproducts and initially explored extraction and detection methods of them.Firstly, we compared six extraction methods of galanthamine and selected hydrothermal refluxing extraction in the end for its higher extracting rate. By orthogonal experiments, it is conformed that the optimal extraction conditions were as follows: running three times with the raw material-95%ethanol ratio at 1/5 and each time lasting for two hours.The solvent extraction and upstream silica gel column chromatography are combined to separate galanthamine. In the former method, the purity is 14.73%, and the recovery is 89.57%. In latter one, the purity can achieve 62.20%, and the recovery is 78.34%.The detection method of galanthamine is established. The wavelength is 290nm, acetonitrile-phosphate buffer (acetonitrile: phosphate=15:85) is used as mobile phase with sensitivity of 0.01AUFS. On this basis, the quantities of galanthamine contenting in different parts of Lycoris radiate Herb. follows such order: bulbs > fibres>leaves, and the end of July>early June>early April.Secondly, we have explored extraction and isolation method of pseudolycorine. Combining with the results of HPLC and UV-spectrophotometry, we initially specula- ted the product is pseudolycorine by salting-out and mixed impregnant method.Finally, we extracted polysaccharide through water hydrothermal refluxing extraction. After eliminating lipid and protein, we separated and purified polysaccharide with DEAE-cellulose column and Sephadex column, the content of final product is 62.51%.
