Objective: To determine the inhibiting effect on serum amylase, TNFa, TXB2 ofsomatostatin for sevre acute pancreatitis (SAP) in rats model and its functionalmechanism.Methods: Sixty four rats were randomly divided into 4 groups, including non-therapeuticgroup,pantreatitis group,control group,and somatostation group,each group havesixteen rats. SAP model was established by means of the method on retrogradeinjection of sodium taurocholate into cholangiopancreatic duct. All SAP groups weretreated by somatostatin.The changes of serum amylase, TNFa, TXB2 and thepathological demonstration of the pancreases and the lung were observed to studytherapeutic effect of somatostatin for SAP model in rats.Results: The experimental results were as follows: (1) The level of serum amylase, TNFa and TXB2 were obviously increased (P<0.01) and obvious pathological changesoccurred in the pancreas and the lung in SAP rats model; (2) The serum amylase,TNFa and TXB2 were obviously increased, the ascites reduced and the pathological changeshave alleviated in the pancreas and the lung by means of somatostatin group comparedwith non-therapeutic group (P<0.05) (3) The inhibiting effect of somatostatin on serumamylase strengthen along with time extending in different period of the second hoursand the fourth hours (P<0.01).The inhibiting effect on TNFαand TXB2 are obvious in 2hours and are not obvious in 4 hours (P<0.01).Conclusion: Somatostatin has obvious inhibiting effect for the release of pancreaticamylase and has obvious inhibiting effect for TNFa and TXB2 in the early course ofSAP but has not the obvious inhibiting effect on TNFa and TXB2 in the terminal courseof SAP.
