Effects Of Yishenyin And Glucosidorum Tripterygll Totorum On Protein Expression Of TNF-α And IL-6 In Rats With Mesangial Proliferative Glomerulonephritis

Objective To observe the therapeutic effects of Yishenyin and Glucosidorum Tripterygll Totorum on mesangial proliferative glomerulonephritis in rats and to investigate the influence of Yishenyi and Glucosidorum Tripterygll Totorum on protein experssion of TNF-a and IL-6 in the rat's kidney of mesangial proliferative glomerulonephritis.Methods 50 normal Wister rats (50days's age) were randomly divided into normal group, model group, Glucosidorum Tripterygll Totorum group, Yishenyin group and common group. The rats in last four groups were established rat models of mesangial proliferative glomerulonephritis respectively. From 5 weeks, Glucosidorum Tripterygll Totorum, Yishenyin and combined Glucosidorum Tripterygll Totorum and Yishenyin were given to the rats in the last three groups respectively. While rats in normal group and model group were given normal water of same quantity as a control. 24 hour urine protein excretion was observed before the experiment and 12 weeks. Varies of biochemistry indexes were observed in 12 weeks when all the rats were executed. Compared with kidney pathological changes in different groups. Meanwhile TNF-a and IL-6 expression in glomerulus was detected by using immunohistochemistry.Results (1) There were not renal tissue pathologic changes in normal group. The pathological manifestations of the model group were enlarged glomeruli, diffuse mesangial proliferation, inflammatory, cell infiltrate mesangial region. In the Glucosidorum Tripterygll Totorum group or Yishenyin group, renal tissue pathological changes were improved than model group, which showed mild or partial proliferation of mesangial cells and substrate slightly, slight inflammatory cell infiltrate mesangial region. After Glucosidorum Tripterygll Totorum and Yishenyin both treated rats with mesangial proliferative glomerulonephritis, renal tissue pathological changes were improved markedly, which showed minimal proliferation of mesangial cells and substrate, few inflammatory cells infiltrate mesangial region. (2) The expressions of TNF-a and IL-6 were sparing in normal rats group. There were only slight TNF-a in the renal proximal convoluted tubule and slight IL-6 in the glomerular mesangium and glomerular capillary. Renal tissue excrete massive TNF-a and IL-6.The expression of TNF-a and IL-6 was increased conspicuously in model group(P<0.05). In the Glucosidorum Tripterygll Totorum group or Yishenyin group, the expression of TNF-a and IL-6 was decreased (P<0.05) than that in the model group. The expression of TNF-a and IL-6 was decreased conspiously in the common group(P<0.01) (3) The levels of urine protein (U P) of 24 hours, blood urea nitrogen (BUN), serum creatinine (Scr), triglyceride(TG), cholesterol(TC) and plasma-albumin(ALB) were all normal in the normal group. In the model group, biochemistry indexes manifested that increased urine protein (U P) of 24 hours (P<0.01), increased triglyceride(TG) and cholesterol(TC) (P<0.05), decreased plasma- albumin (ALB) (P<0.05), normal blood urea nitrogen (BUN) and serum creatinine (Scr) levers(P>0.05). After drug treatment, biochemistry indexes manifested that decreased urine protein (U P) of 24 hours(P<0.05), decreased triglyceride(TG) and cholesterol(TC) (P<0.05), increased plasma- albumin (ALB) (P<0.05), normal blood urea nitrogen (BUN) and serum creatinine (Scr) levers(P>0.05). Those effects were conspious in the common group (P<0.01).Conclusion Yishenyin and Glucosidorum Tripterygll Totorum both reduce the urine protein excretion of rats that with mesangial proliferative glomerulonephritis, rectify dyslipidosis, raise plasma-albumin, decrease the protein expression of TNF-a and IL-6 in glomerulus, and relieve hyperplasy of intercapillary cells and ground substance. Those effects are markedly manifested by using Yishenyin and Glucosidorum Tripterygll Totorum in common.
