Effects Of Shengjiangsan On Expression Of TLR4 And NF-kB In Rats With Mesangial Proliferative Glomerulonephritis

Objective To observe the therapeutic effects of Shengjiangsan on mesangial proliferative glomerulonephritis in rats and to investigate the influence of Shengjiangsan on experssion of TLR4 and NF-kB in rats' kidney of mesangial proliferative glomerulonephritis.Methods 40 normal Wistar rats (50 days old) were randomly divided into contral group, model group, Shengjiangsan group and Lotensin group. The rats in last three groups were made as models of mesangial proliferative glomerulonephritis respectively. From the fifth week, rats in Shengjiangsan group and Lotensin group were feed with water solution of Lotensin and Shengjiangsan decoction respectively. While rats in contral group and model group were feed with normal water of equivalent capacity, and rats can get food and water easily.24 hour urine protein excretion was texted respectively before experiment, the forth week and the12th week. At the 12th week, all the rats were executed and biochemistry indexes were texted. Kidneys were conserved to observe their pathological changes in different groups. Meanwhile TLR4 and NF-kB expression in kidney tissue was detected by immunohistochemistry.Results (1) There were no pathologic changes of glomerulus structure and no inflammatory celles infiltrate in contral group. The pathological manifestations of the model group were enlarged glomeruli, diffuse proliferation of mesangium, inflammatory cell infiltrate in mesangium region. There were mild proliferation in some glomeruli mesangium and its groundmass, but no inflammatory cell in Shengjiangsan group. There were no obviously proliferation in glomeruli mesangium and groundmass in Lotensin group, also no inflammatory cell. (2) The expressions of TLR4 were sparing in contral group, however, there were apparent expressions in glomeruli mesangium, renal interstitium and proximal convoluted tubule of kidney in model group, and expressions of both Shengjiangsan and Lotensin group were weak positive. NF-κB in contral group were only expressed a little in cytoplasm of renal tubular epithelial cell, while there were manifest expressions in both glomerulus and renal interstitium in rats of model group, they are more likely express in nucleolus, but expressions in Shengjiangsan and Lotensin group were weaker. Comparing the expressions of TLR4 and NF-kB of model group with contral group, the model group was higher(P<0.01), Shengjiangsan and Lotensin group was decreased (P<0.05), and there is no difference between this two group (P>0.05). (3) The levels of 24 hours urine protein (UP), blood urea nitrogen (BUN),serum creatinine (Scr), triglyceride(TG), cholesterol(TC) and plasma-albumin(ALB) were all normal in the contral group. In the model group, biochemistry indexes manifested increased UP of 24 hours (P<0.01), increased BUN and Scr levers(P<0.01), increased TC (P<0.05). The TG and ALB had no difference with contral group(P>0.05). After treatment, UP of 24 hours decreased in both Shengjiangsan and Lotensin group(P<0.05), the treatment have no apprent influence to BUN and Scr, only the BUN of Lontinsin group have changed(P<0.01), the treatment have no effect with TG and TC.Conclusion Both Shengjiangsan and Lontinsin can reduce the urine protein excretion of rats with mesangial proliferative glomerulonephritis, decrease the expression of TLR4 and NF-kB in kidney tissue, and relieve hyperplasy of intercapillary cells and ground substance.
