The Experiment al Study Of Zhuang gu Granula On The P ituitary-Adrenal Axis Of Ovariectomized rabbitsObjectives:To investigate the treatmental effect of Zhuanggu Granula on primary osteoporosis,and to study the mechanism from pituitary-adrenal axis.Methods:Osteoporosis model animals(Sprague-Dawley Stain,6 months)by ovary excised were randomly divided into Zhuanggu Granula treating group,Gaierqi group and normal saline group.The animals were killed after 12 weeks treatment in order to collect experimental samples.The levels of TNF-a,COR in serum were measured.The ACTH cells in pituitary were detected by immunocytochemistry and morrphometric observations. Bone mineral density were measured,the undecalcified tibia bones were morphologically observed,the ultrastructural of bone tissue of femur neck were observed by transmission electron microscope.Results:12 weeks after ovariectomy, bone mineral density in model group decreased,the levels of TNF-a and COR were increasing ,the amounts and areas of ACTH cell were more than shamoperative group,the osteocytes became more resorptive and degenerative phase osteocytes by electron microscope. After 12 weeks treatment,bone mineral density increased obviously, the levels of TNF-a and COR decreased, the amounts and areas of ACTH cell were lower, the osteocytes belonged to formative phase osteocytes.The activity of osteoblast was rising.Conclusions:(1)A high turn-over osteoporosis can occur in rats 12 weeks after ovafiectomy. (2) Zhuanggu Granula can effectively treat osteoporosis of ovariectomy habits by accelerating the osteoblastion, enhancing the contents of bone inorgan and improving the properties of trabecular bone. (3)The function of pituitary-adrenal axis of ovariectomy rats were always excessive, which probably relating to the activation of TNF-a to the HPA axis. Zhuanggu Granula can effectively control the pathologic conditions, adjust the function of pituitary-adrenal axis.
