Objective The experimental study was carried out to prove the efficacy of Yigutang in ovariectomized rats and investigated its mechanism.Methods After 12 weeks of operation, female Wistar 40 rats were divided into 4 groups, administered saline after ovariectomy Group A(control), administered Yigutang recipe 1 g/kg after ovariectomy Group B(traditional Chinese drug control), administered α -D3 0.05 μg/kg after ovariectomy Group C(positive control) and administered saline after sham operation Group D(sham-op),and were sacrificed after 12 weeks of Yigutang treatment periods. We examined that Yigutang is capable of affecting osteoblast proliferation using HE Stain and expression of BMP-2 in bone tissue using Immunohistochemistry Stain.Dual energy X-ray absorptiometry (DEXA) was used to measure the bone mineral density (BMD) and bone miniral contents (BMC) . Serum was collected for analysising the level of BGP and CT by applying radioimmunity technique as well as determining the content of Ca and P by using the electrode technique. Bone biomechanical properties test were performed using a material testing machine. Results Compared with Group A, the expression of BMP-2 andcounting of osteoblaster were significantly increased in Group B and Group C (PO.01). The BMD and BMC of Group B, Group C and Group D were significantly increased compared with Group A(P<0.0\). Results showed that the difference between Group B and Group A resulted significant (/><0.05) for CT and BGP,but the serum Ca and P did not have significant difference except Group C.The load that the femur withstood in three-point bending test was also increased in Group B compared with Group A (P<0.0\), and the other biomechanical parameters also inclined.Conclusions Yigutang effectively prevent bone loss and obviously increased the counting of osteoblaster as well as bone biomechanical propertie in OVX rats. High level secretion of CT , BGP and expression of BMP-2 maybe play important roles in Yigutang treatment of osteoporosis.
