Chuanxiong ( Ligusticum Chuangxiong Hort. ), a sort of traditional Chinese medical plant, that there is a centuries-old history of officinal in our country, because of unique officinal value, it is used to treat headache and strengthen the action of circulating blood. This plant distribute extensively in our country, especially in Sichuan, Yunnan, Hubei and Guangxi. In this paper, Chuangxiong was used as study material. Analyzing the structure of polysaccharides from Chuanxiong can provide more theoretical basis for its bioactive.The main experiment result was as follows: a water soluble crude polysaccharide was extracted from dry rhizome of Chuanxiong. LPC was obtained from frozen-thawed, removed protein by the combined deprotionization methods of enzyme/Sevag and Sepharose CL-6B column chromatography. The content of polysaccharide was 98.92%. The molecular weight of it was 4.9×104 Da, which measured by Sepharose CL-6B and HPLC. Gas chromatography (GC) analysis showed that LPC was composing of Glc, Gal , Man, Ara, the molar ratio was 15.56:5.17:3.84:1.00.The structure characteristic of LPC was investigated by partial acid hydrolysis, periodate oxidation, Smith degradation, methylation analysis and IR, GC, GC-MS. The results revealed of methylation that Glc was mainly made up of (1â†'4)-linked, and being little (1â†'6)-linked; Man was mainly made up of (1â†'3,6)-linked and (1â†'3)-linked; Gal was made up of (1â†')-linked, Ara was made up of (1â†'3)-linked. The results revealed of partial acid hydrolysis that the main chain of LPC was mainly made up of Glc and Man, Glc was core of the main chain, Glc, Ara and Man lied to the side chain, the nonreduced end was Gal. The results revealed of that periodate oxidation and Smith degradation that (1â†'4)-linked was mainly link type, and other linkages as (1â†'3,6)-linked , (1â†'3,4)-linked , (1â†'6)-linked, (1â†'3)-linked and (1â†')-linked, which accorded with this result of methylation analysis.A preliminary analysis of the structure of the polysaccharides in Chuanxiong: LPC contained Glc, Man, Gal, Ara. The main chain was composing of Glc and Man, (1â†'4)-linked-Glc and (1â†'6)-linked-Glc were mainly link type; the side chain of LPC composed of (1â†'3)-linked-Ara and (1â†'3)-linked-Man; Residues was substituted at (1â†'3,6)-linked- Man ,and the nonreduced end was Gal .
