ObjectiveAmeloblastoma (AB) is the most frequently occurring odontogenic tumor, the frequency of which is 59.3%. The benign tumor may progressively invade the jaw, recur locally and transform malignantly, therefore, treated as a semi-malignancy tumor, a surprising number of physiological functions are influenced by the phosphorylation/de-phosphorylation-dependent regulation of cellular processes, with many diseases having at their core an underlying defect in intracellular signaling. PP2A is one of the major Ser/Thr phosphatases implicated in the regulation of many cellular processes including regulation of different signal transduction pathways, cell cycle progression, DNAreplication, gene transcription and protein translation. Some studies indicated that this enzyme might actually exert tumor suppressive function.The present study aims to measure the expression of PP2A/PR65 proteinum in AB and to investigate the molecular mechanisms in the regulation of PP2A/PR65; and examine the association between PP2A/PR65 and the clinicopathological characteristics of the tumor specimens in AB to provide some theoretic basis for the diagnosis, therapy and prognosis of AB.Materials and Methdods1. Immunohistochemical staining: A total of 68 AB, 6 ameloblastic carcinomas and 7 normal oral mucosas were collected. I use the immunohistochemical staining S-P to observe the change of the PP2A/PR65.2. Western Blot: To measure the expression of PP2A/PR65 proteinum in 15 AB and 2 normal oral mucosas respectively.Result1. the express of PP2A/PR65 are less in AB.2.PP2A/PR65 involved in the occurrence and development of the AB.
