Bone morphogentic proteins ( BMPs) , a kind of the cytokines which be-longs to the TGF -β cytokine superfamily, play an important role in the cell growth, differentiation, apoptosis, as well as diversified histologic and organic morphogenesis. Some researches have shown that BMP -2 is an important signal molecule in the interaction between the epithelium and mesenchyme during embryonic development. BMP - 2 expression might always been seen in the epithe-lial tissue of eyes, kidney, ureter, lung, small intestine, and prostate throughout the embryonic development. BMP - 2 also plays a role in the dental germ development, odontoblast differentiation, and repair of dental pulp injury. BMP -2 acts not just only in the body growth and disease development, but also express in the case of many body neoplasm such as osteosarcoma, fibrous dysplasi-a, and neoplasm of salivary gland, and it affects their clinic manifestation and prognosis. At present, the molecular biological mechanism of BMP -2 abnormity and the cause of biological behavioral change of the epithelial tumors induced by BMP - 2 are not very clear.Ameloblastoma ( AB) is one of the most common odontogenic tumors with some special characters such as infiltrative growth locally, highly relapse, can-ceration, and metastasis etc. Therefore, some scholars classified Ameloblastoma into the borderline tumor. This experiment planned to take the immunohisto-chemistry method to detect the expression of BMP - 2 in Ameloblastoma. And to perform relative correlations analysis. Study on the relationship between cell differentiation and biological behavioral of Ameloblastoma. Discuss the mechanism of onset and relapse of Ameloblastoma so as to provide the theoretic basis on diagnosis , treatment, and prognosis of tumors.Material and methodsExperiment method: immunohistochemistric detection on expression of BMP -2, with SABC method applied. The specimens are archived wax slices from the 1" Affiliated College of China Medical University and Shenyang Stomatologic Hospital, among of which 34 (18 follicular, 10 plexiform, and 6 parietal; 6 one year history, 21 one to three years history, 7 over three years history) are Ameloblastoma through pathological diagnosis and 14 are of normal oral mucosa. Specimen number of maxillary and mandible are not limited.Result determination: brown yellow granule appearing in cytoplasm means positive coloration. Performing semi - quantitative analysis on the expression of BMP - 2 with modified scoring method. A ( score based on the cellular coloration and shade of colors on slice) : no coloration scores 0; buff scores 1; brown yellow scores 2; chocolate brown scores 3. B ( score based on the percent of col-orated oncocyte to all the tumor cells on slice) : below 25% scores 1; 25 -75% scores 2; above 75% scores 3. Score of each slice = A * B. The resulting groups classified according to scores are: negative ( - ) , which scores 0; weak-ly positive ( + ) , which scores 1 - 4; strongly positive ( + + ) , which scores more than 4.Statistical method: analyzing with SPSS 11.5 statistical software system. Wilcoxon test and Kruskal - Wallis test were used to analyze the data, P <0. 05 means of statistical significance difference.Results1 case among the group with normal oral mucosa appears weakly positive, and the positive ratio is 7. 1%. Total positive ratio in the group of Ameloblastoma is 97.1% , 15 cases are weakly positive and 18 cases are strongly positive. In the case of expression of BMP - 2 in Ameloblastoma positive reaction occurs within the cytoplasm of tumor cells. In the Ameloblastoma with positive reaction positive expression occurs in the columnar epithelium and stellate reticulumcells. Significant difference (P<0.01) can be seen between the expression in the Ameloblastoma and that of normal epithelium, and this intergroup compari-son is of statistical significance.Ameloblastoma can be grouped as follicular AB group, plexiform AB group, and parietal AB group according to the histopathologic type.
