| Xinjiang Dryopteris filix-mas(L.) Schott,a folk medicinal plant which is widely distributed in Xingjiang,belongs to the genus of Dryoperis Adans of Dryoperidaceae family. It has a long history as bugs-killer medicine. From earlier 1750 ,it has already been used for this purpose in Europe,and then it began to widely produced from 1825.The plant has been embodied in pharmacopoeia of England,Germany,Switzerland,Japan and America as bugs-killer medicine against tapeworm and hookworm.So far,there is little research on the flavonoids of Dryopteris filix-mas(L.) Schott, The present study focused on the total flavonoids of separation,purification and its free radicals Scavenging.The effects of different extraction ways such as hot water,the ethanol,ultrasonic and Soxhlet extraction on yield of flavonoids from Dryopteris filix-mas(L.) Schott were studied and finally the technical conditions of ultrasonic extraction was determined.The result showed that the highest extraction rate was 5.53% by Soxhlet extraction,and ultrasonic extraction ranked second.However,because ultrasonic way is ready,labor and material saving and without destruction to effective component of sample,it is choosed in the study.The technical conditions of ultrasonic extraction were optimized as:temperature 60℃, ratio of material to solvent 1:20,extraction for 4 times and 50 min for each time,ethanol 80%.The isolation ability of AB-8 macroporous adsorption resin for total flavonoids of Dryopteris filix-mas(L.) Schott was also studied by examining the adsorption and desorption,and the related parameters were investigated.The results showed that the AB-8 macroporous adsorption resin was optimal for purifying total flavonoids from Dryopteris filix-mas(L.) Schott and its adsorption capacity was determined to be 25.53 mg/g,its desorption rate was up to 98.3%.The pH value of loading sample significantly affect the adsorption properties of resin.The optimum refine technical conditions were as follows:pH value of solution,4.08;the current velocity of adsorption,0.5mL/min;the elution velocity,1.0mL/min; the elution reagent,1.2BV 30% and 1BV 50% ethanol.The total flavonoids was relative stable under certain sunlight strength,moderate temperature,mild acid solution and low concentration saccharose solution,but was unstable in alkaline or saline solution.The different solvent extraction of total flavonoids had good free radicals scavenging capacity against·0H and 02-·,especially scavenging of·0H by acetidin extraction whose scavenging rate was up to 99.5% which was stronger than that of VC, 90.2%.The different solvent extraction also had favorable inhibitory effect against lipid peroxidization,and had good stabilization. |
