Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.(Aquifoliaceae)vrieties of the genus Ilex pubescens Hook.et Arn.var.Which is mainly distributed in the western southern Guizhou(Libo),Guangxi(Baise,Lingyun)and southeast Yunnan(Funing,Yanshan),born in the altitude of 550-1410 meters of evergreen broad-leaved forest.The modern pharmacological researches have shown that this plant had many efficacies,such as anti-oxidation,anti-inflammatory,neuroprotective activity,anti-tumor activity,antiviral effect and other pharmacological effects.In addition,Modern phytochemical studies have shown that this plant mainly contains triterpenoids and saponins compounds,flavonoids and their glycosides,alkaloids,terpenoids and other chemical constituent.At present,there are many reports on the research of Ilex pubescens Hook.et Arn.var.,but there is no report on the research of this plant variant,Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.whose material basis and pharmacodynamics research is not clear.Based on the review of the chemical constituents and pharmacological activities of the genus Ilex L.,the chemical constituents of the roots of Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.collected from Lingyun County of Guangxi Province were systematically studied.As a result,using various separation techniques,such as extraction,vacuum silica gel column chromatography,medium pressure rapid preparative chromatography,high pressure rapid preparative chromatography,Sephadex LH-20,recrystallization and semi-preparative liquid chromatography,and their structures were elucidated by chemical properties and spectromatography analysis(NMR,MS),35 compounds were isolated,and 33 compounds were identified from the ethyl acetate fraction of Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.Their structures were identified to be oleuropein(MD-1)? oleoside 11-methyl ester(MD-2)?(8E)-nuezhenide(MD-3)?(8Z)-ligstroside(MD-4)?oleoacteoside(MD-5)?oleoside dimethyl ester(MD-6)?(2??R)-2??-methoxyoleuropein(MD-7)?(2??S)-2??-methoxyoleuropein(MD-8)?butyl isoligustroside(MD-9)?liguside B(MD-10)?ligustroside(MD-11)?isoligustroside(MD-12)?oleonuezhenide(MD-13)?oleuropein-3''-methyl ether(MD-14)?jasurolignoside(MD-15)? olivil-4'-O-?-D-glucoside(MD-16)?(+)-cyclo-olivil 6-O-?-D-glucoside(MD-17)? wilfordiol B(MD-18)?(-)-olivil(MD-19)?(erythro,erythro)-1-[4-[2-Hydroxy-2-(4-hydroxy-3-methoxyphenyl)-1-(hydroxymethyl)eth oxy]-3,5-dimethoxyphenyl]-1,2,3-propanetriol(MD-23)?pinnatifidanin B II(MD-20)?prinsepiol-4-O-?-D-glucopyranoside(MD-21)?sinenoside I(MD-22)?hebeoside(MD-24)?3,4-dihydroxyphenyl ethanol(MD-25)?salidroside(MD-26)?acteoside(MD-27)?isoacteoside(MD-28)?torenoside B(MD-29)?sinapic aldehyde 4-O-?-D-glucopyranoside(MD-30)?(Z)-sinapic acid-4-O-?-D-allopyranoside(MD-31)?syringin(MD-32)?vanillyl-?-D–glucopyranoside(MD-33).Among them,MD-6~MD-15 ? MD-18 ?MD-20~MD-29?MD-30?MD-31 and MD-33 were isolated from this genus for the frist time.The other compounds were isolated from Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.for the frist time.In this paper,the chemical constituents of Ilex pubescens Hook.et Arn.var.kwangsiensis Hand.-Mazz.were studied systematically,and 33 compounds were isolated to provide the material basis for the further study of the pharmacological research and provide the material basis and theoretical basis for the further development of the plant.
