Background and Objects:Alzheimer's Disease, a neurodegenerative disease which develops with age, is characterized by evident learning and memory impairment and dementia, and it's etiopathogenisis is still unknown.It's disease incidence increases by age, the morbidity rate is 5% w. ith the age above the age of 65 years old, compared with 20% above the age of 80 years old.The pathology characteristics are the deposition outside the brain cell of senile plaque (SP) which center isβ-amyloid protein(I.e. Aβ) and the absence of neuron caused by neurofibrillary tangles inside the neuron and regional neurocyte decease originated by neurocyte Apoptosis inside the brain. There are many Chinese crude drugs which are good for .the therapy in TCM. this text approach Nao-ling Decoction take part in the changes of AD model rats' learning, memory and pathological morphology ,and the effect on TGFβ1 and TNFα, Which lays the theoretical foundation for further elucidating its mechanism and treatments for AD.Methods:There were 40 pure line SD rats, then they were divided in random into a Nao-ling Decoction group (that was traditional chinese medicine group, TCM group),a.Western medicine group,a model group,a sham operation group and a control group. Each group had 10 rats.The AD model was established by injectingβ-amyloid protein 1-42(Aβ1-42) into the dorsal of double sides hippocampus. feeded a week before the operation,they were cured for twenty-eight days in all.then, their ability of leaming and memory was detected by Y-maze and Morris water maze test,and the morphology of CA3 hippocampus was shown by HE stain.and the neuron apoptosis was shown by Nissl stain.the expression of TGFβ1 and TNFαin the hippocampus was measured with immunohistochemical stain.Results:1. Y-maze test: The times needed to learn how to evade the electric stimulation in the model rats were significantly more than those in the TCM group(P<0.05) and Westem medicine group(P<0.05). Morris water maze test: Compared with normal group, the escaped and latent period of model group was delayed significantly in Morris water maze scoured experiment on delitescence platform (P<0.05); In exploratory experiment, the average pass through times in sham operation group and normal group were more than that of other three ones, and that of the model group was lest, there were significant difference (P<0.05).2. Three stains: HE stain: In AD model rats group, the number of neuron in hippocampus CA3 sector was less and the array of neuron was more irregular than those in TCM group and Western medicine group. The TCM group was better than the Western medicine group; Nissl stain: In AD model rats group there were less tigroid bodies in pyramidal cell and more amounts of pyknosis and trachychromatic caryons compared with TCM group and Western medicine group; Congo red stain: there was deposition of.β-amyloid protein in model rats group hippocampus sector, while there was obviously less in TCM group and Western medicine group.3. The compasion of masculine cells about TGFβ1 in each group, the model one was lest and TCM group, Western medicine group were more than that of the other ones (P<0.05), so did the IOD value (P<0.05). They had statistical significance, while the inflammatory factor TNFα,the model one expressed most and TCM group, Western medicine group were less than that of the other ones (P<0.05), so did the IOD value (P<0.05).Conclusions:1.The AD model prepared by the injection of Aβ1-42 into the dorsal of right hippocampus is reliable and reproducible.2.The learning and memory of the model rats were decreased obviously, Nao-ling Decoction can improve the learning and memory of the model rats(P<0.05).3. Nao-ling Decoction can enhance masculine cells and IOD value of TGFβ1 in hippocamp sector and can protect pyramidal cells of hippocamp in AD rats. While decrease the expression and the IOD value of the inflammatory factor TNFα, accordingly lessen the deposition ofβ-amyloid protein in AD rats hippocamp.
