Propolis is a natural sticky resinous substance collected by honeybees fromcracks in the bark of trees and leaf buds enriched with bees'salivary enzymes andwax.As a folk medicine, it had been reported to possess various biological activitiessuch as antibacterial, anti-inflammatory, antifungal, antiviral, enhancing the immuneregulation, anticancer, antioxidant, scavenging free radicals, anti-radiation, promotingtissue regeneration, antilipemic, etal.In recent years, propolis was extensively noticedover worldwide for it's strong health-maintain and adjuvant-therapy functions tohuman beings. The thesis focused on the extraction and separation of fat-solubecompositions ofpropolis, the analysis of it's chemical compounds and the trial-prod-ucing of the new-style propolis capsule, the results were as follows:Organic solvent and super critical CO2 were adopted to extract the activecompositions of propolis. The best conditions for processing organic solvent extracti-on at a labolatory scale were temperature at 70℃and time for 8 hours; proportion ofpropolis and solvent was 1:6.5; the extraction rate of the active compositions was62.5%. The best processing conditions of super critical CO2 for the dewaxed propolisextraction at a pilot scale were: operation pressure 30Mpa; operation temperature55℃; operation time 240mins; CO2 flow rate 200kg/h;the extraction rate of theactive compositions ranged from 15.5%to 21.5%.The dewaxed ethanol insoluble partof propolis was extracted by super critical CO2 under the same condition withdewaxed propolis, the extraction rate of the active compositions ranged from 33.75%to 46.25%. The propolis resource could be utilized sufficiently with the technologycombined organically by the organic solvent and super critical CO2 extractionthrough experiment.The total flavanoids in SCF-CO2 extract from propolis and it's ethanol insolublepart were determined respectively by HPLC and Spectrophotometry, the results were6736.78 mg/100g and 2903.44 mg/100g by HPLC and 6900.44mg/100g and2903.43mg/100g by Spectrophotometry.It showed that the results obtained by thosetwo method are basically similar. The organic compounds of SCF-CO2 extract from propolis and it's ethanolinsoluble part were identified and quantified by GC/MS,the results showed that therewere 5 kinds of flavanoids, 12 kinds of terpenoids and 29 kinds of organic acids with arelative content of 14.3%,7.24%, 19.64%in the former, 3 kinds of flavanoids, 13 kindsof terpenoids and 10 kinds of organic acids with a relative content of 5.02%,1.56%and 10.86%in the latter.The content of Pb element in the SCF-CO2 extract of propolis was 0.17mg/kgdetermined by ICP/MS,which was lower than about 1/5 of that of the nationalstandard.The active compositions in the SCF-CO2 extract of propolis were separated bythe organic solvents and silica chromatography, the results showed that the mainactive compositions such as flavonoids and terpenoids were accumulated in theelution of ethyl acetate during the silica chromatography experiment.The hydroxyl free radical(·OH) and superoxide anion free radical (·O2-) scaven-ging ability of EEP was stronger than that of the SCF-CO2 extract of propolis.Thebacteriostasis ability of SCF-CO2 extract of propolis was overall less stronger thanthat of the EEP, but far more stronger than that of the WER The SCF-CO2 extract ofpropolis had an outstanding inhibition ability of E. coli that the EEP did't have.The inclusion of the new-style propolis soft capsule was composed by SCF-CO2extract of propolis, octacosanol, soybean lecithin and refined edible oil at a ratio of4:1:1:5 by weight.The capsule's shell was formed by the combination of gelatin andglycerol at a raito of 1:0.35,the soft capsule had a good quality of stability.
