Objective:To investigate the expression of MMP-10 in human non-small cell lung cancer(NSCLC)and to evaluate how the indicator relate to patients'sex,age,pathological type,clinical TNM stage and metastasis in lymph node,and to investigate the correlation between the expression of MMP-10 and the expression of CK19 mRNA in peripheral biood of NSCLS patients.Methods:In the present study,it is used the real-time fluorescent quantitative reverse transcriptase-polymerase chain reaction(real-time PCR)and the immunohistochemical method.Before preparing of preoperation,36 NSCLC patients were collected peripheral blood samples with penetration of superficial antebrachial vein and their NSCLC tissue sample and para-cancerous normal tissue were used.11 healthy people and 19 patients being lung disease served as control,who were collected peripheral blood samples likewise.Those patients being lung disease hadn't undergone any examination such as penetration and bronchofibroscopy. NSCLC cancerous tissue and para-cancerous normal lung tissue were examined the expression MMP-10 with the immunohistochemical method.Using cytokeratin19(CK19)as a marker CK19 mRNA's expression of all peripheral blood samples was measured by real-tine PCR.The data of MMP-10'sexpression is recorded as '+.-',based on the percentage of cells in positive expression of MMP-10,<25%as(-),25-50%as(+),50-70%as(++),>75%as(+++).The data of CK19 mRNA in peripheral blood samples was got relatively quantitatively from formular, which is Nck19=2Ctck-CtGAPDH,and the expression of GAPDH mRNA is the inner control.All data was statistically analysed with SPSS 12.0 softward for windows.Results:The expression of MMP-10 was located in cytoplasm.The expression rate of MMP-10 in NSCLC was 44.4%(16/36).There wasn't expression in the para-cancerous normal lung tissue. The relation between the expression of MMP-10 and clinicopathoigical character:the expression level of MMP-10 in NSCLC was significantly correlated with the pathological grading(Gamma Value=0.685,Fisher' exact test P=0.002,P<0.01),the worse the pathological grading was,the conspicuous the expression was.The positive expression of MMP-10 was associated with clinical TNM stage(Gamma Value=0.584,Fisher' exact test P=0.008,P<0.01),evident expression of MMP-10 showed advanced clinical stage.Expression of MMP-10 in NSCLC tissue with lymph node metastasis was higher than without this kind of metastasis(Fisher' exact test x2=9.706 P=0.014 P<0.05).positive expression rates of MMP-10 in NSCLC have no link with pathological type(adenocarcinoma or squamous cell carcinoma),age,sex(P>0.05).The expression of MMP-10 inner NSCLC tissue had positive correlation with high expression quantity of CK19 mRNA in peripheral venous blood(γs=0.796,P<0.01),and this kind of expression on the high level indicated the existence of the circulating tumor cells and the probability of micro-metastasis.The data of CK19 mRNA in peripheral venous blood of healthy people and the patients being lung disease,which was gave by real-time PCR,indicated that there was a low level discovery of CK19 mRNA in peripheral venous blood of non-carcinoma people,which was got with superficial antebrachial vein penetrated.The latter resulted from clinical normal factors and was essentially different with the former(Z良性vs患者=-5.666 P<0.01; Z健康vs患者=-4.322 P<0.01).Conclusions:1 There was a high ratio of expression of MMP-10 in NSCLC tissue.MM-10 wasn't expressed in para-cancerous normal lung tissue.2 The expression level of in NSCLC tissue was significantly positively correlated with the pathological grading,the clinical TNM stage,the metastasis of lymph node and the expression of CK19 mRNA in peripheral vein blood.3 The examination of MMP-10'sexpression in NSCLC tissue may be a great help to understand the tumor's malignant behavior to assess patient's prognosis to furnish reasonable and necessary therapy.
