Objective: To study the effect of Jiayijian in treatment of invasion and metastasis in mice with transplanted liver cacer(H22),especially to observe the effect of Jiayijian on Matrix metalloproteinases on mice transplanted hepatocarcinoma,so as to make sure the mechanism of anti-invasion and metastasis of Jiayijian on the mice hepatocellular carcinoma,to provide foundation for the clinic appliance of Jiayijian.Methods: 75 male mice (SPF) were transplanted liver cancer(H22) sudcutaneously into left footpads to be established as experimental lymphatic metastasis models, and were divided randomly into 5 groups(each group had 15 mice): the pathologic model group(group A), Jiayijian low, middle, high dose treatment groups ( group B,C,D),the western medicine group(group E). From the next day, the groups were treated by Jiayijian low, middle, high dose and 5-Fu respectively, the pathological group was given the same dose NS(Normal Saline),Once everyday. After 20 days, the mice were killed and weighed. The transplanted lateral popliteal fossa, para-iliac artery and renal lymph nodes, thymuses, spleens, and their left footpads were excavated, weighed and some parts of them were performed on parafomaldehyde-fixed, paraffin-embedded sections. Count the index of lymph nodes, thymuses, spleens. The invasion and metastasis occurring in footpads and lymph nodes were observed and computed, the expression of Matrix metalloproteinases were also measured with immunohistochmeistry staining.Results: Compard with pathological model control group, the Jiayijian treatment groups can decrease the level of invasion and metastasis in footpads and popliteal fossa lymph nodes of hepatocellular carcinoma, the index of lymph, and the low dose group (group B)were significantly decreased(P<0.05). The Jiayijian middle, high dose groups (group C,D)can significantly increase index of thymus(P<0.05), the western medicine control group( group E)decreased the index of spleen(P<0.05), while the Jiayijian groups didn't have the side-effect(P>0.05). On the other hand, the group B, C, D can significantly decrease the Matrix metalloproteinases.Conclusion1 The Jiayijian can inhibit the invasion of the hepatocellular carcinoma in mice's footpads and lymph nodes, and it can decrease the index of lymph node, prevent lymphatic metastasis.2 Its mechanism is to improve immune functions and injury the carcinoma cell, to decrease Matrix metalloproteinases in liver cancer, so as to inhibit the tumor angiogenesis. And the effect can be improved with the increase of Jiayijian dose in proper range.
