ObjectivesTo observe the effect on the function of reproduction and the morphologicalchanges of the neurons,astrocytcs and myelin basic protein in hippocampus ,thepyramidal cells in internal pyramidal layer of the cerebral cortex motor area in adultBalb/c mice after fed with different dosage KI water for 3 months and 6months. Tostudy the influence of iodine deficiency or iodine excess on the function ofreproduction and the morphology and functions of Cerebral cortex and hippocampus.MethodsOne month Balb/c mice, half of male and half of female, were divided into fivegroups randomly:low iodine group (LI group), normal iodine group (NIgroup), 5, 10, and 50 folds of normal normal iodine intake groups (5HI, 10HI, and 50HIgroups). All mice were sacrificed at the end of the third and the six month respectivelyapart from selecting out female and male were mated by 3:1 after fed three month andobserve the fertility of female mouse. then about 10 mice were selected randomlyfrom each group by us to observe the morphological changes of the hippocampus andcerebral. The expressions of NSE in the neurons of the hippocampus CA3 region, theexpression of GFAP in the hippocampal astrocytes were examined byimmunohistochemistry, and volume density (Vv), numerical density on area (NA) andthe Average optical density (AOD) together with the Average optical density of MBPwere measured.The pyramidal cells in internal pyramidal layer of the cerebral cortex motor area werealso examined by silver impregnation technique, the dendrites arising from basalportions of the pyramidal cells were counted. The area percentages of the molecularlayer were measured after strained by the special purple dye.Results1. The function of reproduction: The conception rate and the number of filial increased in filial generation than the control. there is no difference between thecontrol and 5HI group. In 10HI there is the conception rate and death rate of pregnantmouse higher than the control. In 50HI, the conception rate and the death rate offilial was lower than the control as well. but there is no different in the number offilial generation between the control and 50HI group.2. The morphological changes of the hippocampus and cerebral:Compared withNI group, the change in morphological of the pyramidal cells in internal pyramidallayer of the cerebral cortex motor area by silver impregnation technique and theimmunohistochemistry in the neurons of the hippocampus in LI group and all iodineexcess groups were no insignificant at 3 month and 6 month.3. The function of cerebral and hippocampus: Both iodine deficiency and iodineexcess will probably make the energy metabolism of the cerebral neurons lowercompared with the control, but the LI more serious.ConclusionsIodine deficiency or iodine excess can affect the function of the reproduction inmice but the iodine deficiency more serious. Meanwhile, iodine deficiency or iodineexcess can not make significant morphological change of the neurons,the astrocytesand the myelin basic protein in hippocampus, of the pyramidal cells in internalpyramidal layer of the cerebral cortex motor area in adult Balb/c mice. But iodinedeficiency or the concentration of iodine much more than normal intake can lead tothe synthesis function of the thyroid low, as a result, the concentrations of T4 and T3decrease. therefore, the energy metabolism of the cerebral will lower. Both iodinedeficiency and iodine excess (more than 50 times of normal intake) can affect on thefunction of the cerebral neurons.
