Objective: To investigate the immune modulatory effect of the Caulerpa racemosavar peltata polysaccharides and selenizing product (CRVPESP-nano-Se) on immuneorgan index, T lymphocyte subgroups and NK cells in model mice.Methods: The first step: the sulphated polysaccharides from Caulerpa racemosa varpeltata (Lamx.) Eubank were extracted by the method of refluxing hot water. Somephysicochemical property of this crude polysaccharides was analyzed and molecularweight was also determined by GPC. The second step: the liquid phase system wasprepared, sulphated polysaccharides (or sucrose) in the use of nano-crystallization andstabilization for surface-modifying nano-Se, that managed to combine CRVPESP (orsucrose) and nano-Se stably. The morphological character and particle diameter ofCRVPESP and nano-Se were observed with scanning electron microscope, trans-mission electron microscope and laser light scattering image. The third step: themodel mice were injected hydrocortisone, it result in weak immune function. Thepositive drug control was levamisole. After intragastric administration liquor ofSodium Chloride, CRVPESP, sucrose-nano-Se, CRVPESP-nano-Se 10 daysseparately, the blood immunity parameter (CD3+, CD4+, CD8+ T lymphocytesubgroups percentage) and the NK cells percentage from spleen were detected withflow cytometry, the thymus index and spleen index of each group were alsomeasured.Results: (1) The sulphated contents of CRVPESP is 30.90%, it's a heteropoly-saccharide, which made of glucose and galactose mainly. It is rich in kinds ofelements, such as Ca, Mg, Al and Fe. Its molecular weight is Mn 3990, Mw 8478. (2)The CRVPESP could modulate the reaction rate, modify the surface of nano-Se andcontrol the assembly of nano-Se. Observed with SEM and TEM, the CRVPESPappeared cotton-like shape and the nano-Se is regular sphere. In addition, the nano-Sewas well distributed in CRVPESP aqueous solution; it could be adsorbed orenveloped by CRVPESP. (3) After intragastric administration liquor of CRVPESP, sucrose-nano-Se and CRVPESP-nano-Se, the significant differences to model groupwere found, the marked upgrade in percentage of CD3+, CD3+CD4+, NK cells andCD4+/CD8+(P<0.05) were concomitant with the obvious increase of thymus index (P<0.05). The immune modulatory effect of CRVPESP-nano-Se on CD3+, CD3+CD4+cells in model mice was better than CRVPESP, sucrose-nano-Se and levamisole.Conclusion: The immunologic data indicated that the CRVPESP could modulate thequantity percentage of T lymphocyte subgroups and NK cells positively in modelmice. The polysaccharides and selenizing product combined CRVPESP with nano-Se,which effected CD3+ and CD3+CD4+ cells significantly.
