Objective To study the therapeutic effects of radix astragali(RA) and safflor injection onbrain-derived neurotrophic factor(BDNF), neurotrophin-3(NT-3) and neurotrophin-4(NT-4) in ratsbrains following local cerebral ischemia/reperfusion.Methods One hundred eighty male adult SD rats were randomly divided into five groups: thesham-operated group, the model group,the RA group,the RA and safflor group. The model of middlecerebral artery occlusion(MCAO) was established by thread ligation method, and at 12h, 30min beforeoperation, rats were injected intraperitoneally corresponding medicine in different groups and themedicine was injected after operation every other 12h. In sham-operated group and the model group,equally dose saline was injected intraperitoneally. Neurological system symptoms were evaluatedafter 6h, 12h,24h,48h of ischemia/ reperfusion, Then the rats were decapitated at 6h,12h,24h,and 48hafter reperfusion, and rats brains were taken for immunohistochemistry and histopathologicalexaminations, positive immune reacted cells of BDNF, NT-3 and NT-4 are counted under lightmicroscope at different time points of ischemia/reperfusion.Results RA and safflor injection can significantly alleviate neurological system damage symptoms(p<0.05); Histopathological examination showd that the cerebral ischemic damage in the RA andsafflor group was significantly milder than that in the model group. and positive immune reacted cellamounts of BDNF, NT-3 and NT-4 were greatly enhanced in high-dose RA group as compared withthe model group and the FC group (p<0.05)Conclusions The protective effects of the RA and safflor injection therapy against cerebralischemia/reperfusion injury might be associated with improving the positive expressions of BDNF,NT-3 and NT-4. Moreover, The therapeutic effects of high-dose RA is superior than the ones of safflorand RA.
