Rodgersia aesculifolia Batal. is the dry rhizomes of Saxifraceae plant-genus Rodgersia Gray. As a traditional Chinese medicine for hundreds years, theRodgersia Gray. is an effective broad-spectrum antibacterial and antiviral medicine forthe treatment of rheumatoid arthritis, chronic bronchitis, dysentery, asthma bronchial,inflammation and chronic gastritis. The volatile components from the dry rhizomes ofRodgersia aesculifolia Batal. is extracted in this work, using Soxhlet, ultrasonic-steamdistillation, ultrasonic assisted extraction, supercritical fluid extraction and microwaveassisted extraction, respectively. The extracts obtained by above extraction methodswere analyzed by GC-MS and the results were compared with each other.There are two Parts in this thesis:Part one: ReviewRodgersia aesculifolia Batal. is the dry rhizomes of Saxifraceae plant-genusRodgersia Gray., and a folk medicine with widely therapeutic range. Modernpharmacological studies demonstrated that it have a certain effect on several sides suchas promoting blood flow to regulate menstruation, getting rid of rheumatism, clearingheat antitoxicant, arresting bleeding, antidiarrheal agent, antivirus action, antimicrobialactivity and immune enhancement. In addition, clinical research have indicated it hasefficacy in the treatment of dysentery, diarrhea, chronic bronchitis, chronic gastritis,thyroid diseases and rheumatic arthritis. With Traditional Chinese medicinemodernization the pharmaceutical action and chemical constituents studies aboutRodgersia aesculifolia Batal. have been attracted attention increasingly. In this thesis,volatile components were extracted by several extraction methods from Rodgersiaaesculifolia Batal. rhizomes and analyzed by gas chromatography/mass spectrometry.Chromatography, with a lot of advantages such as high separation capability, highsensitivity and short time analysis, plays an important role in analyzing complexmixture sample and essential oil as a first-choiced method. Due to its shortcoming inqualitative analyzing, chromatography just acts as an assistant technique for qualitativeanalysis. Spectral analysis methods, such as Mass Spectrogram (MS), InfraredSpectroscopy (IR) and Nuclear Magnetic Resonance (NMR), have very strong ability of structure identification, but don't have separation capability. So they can not be used fordirectly analysis of complex mixture. The technique of chromatography coupled withspectral analysis becomes an effective technique for analysis complex mixture. Atpresent chromatography has been realized to combine with all of the usual spectralmethods. Gas chromatography-mass spectrometry, as far as gas chromatography isconcerned, is a method of most widely used and successful application.The extraction methods of natural organic compounds were classified intotraditional extraction methods and modern extraction methods. Traditional extractionprocedures are time-consuming, however, and some suffer from losses of some volatilecompounds, low extraction efficiency, and toxic solvent residue in the extract. Thesedisadvantages have led to the consideration of new techniques which can be filled withthe development for Traditional Chinese Medicine Modernization. Modern extractionmethods are based on advanced modern apparatus or advanced technology, and havebeen recently acted as an alternative method in some ways. This thesis summarizes theprinciple of each extraction methods, extraction apparatus, extraction procedure and itsapplication on the active component extraction from natural products.Part two: Research papersIn this part, it includes three main contents:First, the extracts from Rodgersia aesculifolia Batal. were analyzed by GC-MS, andthe determination condition was optimized at the aspects of chromatographic column,injection temperature, split ratio, temperature programmed, and so on.Second, the volatile componet of the dry rhizomes of Rodgersia aesculifolia Batal.is extracted, using Soxhlet, ultrasonic-steam distillation, ultrasonic assisted extraction,supercritical fluid extraction and microwave assisted extraction, respectively.Third, The extracts from Rodgersia aesculifolia Batal. obtained by the aboveextraction methods were identified respectively under the optimum GC-MSdetermination condition. And the results analyzed by GC-MS were compared.
