Objective:Through blood samples were taken from rats with hypertriglyceridemic severe pancreatitis and severe acute pancreatitis, to measure the concentrations of triglyceridemia,endothein-1,nitric oxide,Interleukin-6,Tumor necrosis factor alpha and the gradement of the pathology severity of pancreatitis.To explore the relationship between hypertriglyceridemic concentration and the pathology severity of pancreatitis. the influence of hypertriglyceridemia on serum IL-6,TNF-a, NO and ET-1 in rats with severe acute pancreatitis.Methods:54 SD rats were used in all experiments.and divided into 3 groups randomly,every group have 18 rats: group A (HLSAP group):feeded by hyper-fat fodder then became servere acute pancreatitis; group B (SAP group):feeded by general fodder,then became severe acute pancreatitis ; group C (sham operation group):feeded by hyper-fat fodder then had a sham operation. hyperlipidemia was induced by feeded with hyper-fat fodder for three weeks,acute severe pancreatitis was induced by retrograde injection of 0.1ml/0.1kg body weight of 5% na-taurocholate sure of 30 cm H2o,as previously described by other authers in group A and group B. then.three-groups were killed in 4 hours,8 hours, 12 hours after the modle had been mad . Blood sample were taken from all subjects to measure triglyceride ,amylase, ET-1 (determined by radio immunoassay), NO,TNF-a and IL-6,..pancreatic tissue samples were taken from head of pancreas and dyed by HE,the degree of pancreatic damage was judged microscopically according to the point score of Schmidt and Pozsar's methods..Results:1. In the animals with no-hyperlipidemia and severe acute pancreatitis have significantly higher(P<0.05) in hyperlipaemia after operation,but had no significant chang(p>0.05) occurred in their three periods.2.In the animals with hyperlipidemia and severe acute pancreatitis developed significantly higher(p<0.05) ET-1 than the animals of the non-hyperlipamic severe acute pancreatit group in 4 hours and 8 hours period,But between group A and group B there was no significant changes(p>0.05) in NO; ET-1,NO declined in group A and group B at 12 hours period, there was no significant changes in ET-1, But group A have significantly higher(P<0.05) decline than group B in NO.3. The concentration of IL-6,TNF-a in group A have significantly higher(P<0.05) than that in group B in all periods (4,8,12hours)4. There are significant correlation between the histological degree of pancreatic damage and the serum triglyceridemic concentration(r=0.542).It can be observed pancreas oedematous ,engorgement, necrotizing,ascites in group A and group B after severe acute pancreatitis had been induced. the histological degree of pancreatic damage were significantly higher in group A than in the group B at all times.Conclusion:1.Mircrocirculation disorder had exsited in the early of SAP.hypertriglyceridemia can improved ET-1and NO rising more earlier and higher in severe acute pancreatitis ,then declining in the late.2.Pancreatitis can caused the hoist of TG once a time. hypertriglyceridemia intensifies the pathologic-histological degree of pancreatic damage . 3. hyperlipidemia may intensifies the expression of IL-6,TNF-a in rats with severe acute pancreatitis.
