Expression And Significance Of CD147, MMP-9 And TIMP-1 In Cervical Lesions

Objective: To detect the expression of CD147,MMP-9 and TIMP-1 in normal cervical epithelial tissue ,cervical intraepithelial neoplasia (CIN) and squamou-cell carcinoma of uterine cervix, and to investigate the relationship of those indexes and the genesis and development of cervical lesion, and to determine whether there was some association in those indexes, by which to look for molecular marker of uterine cervix cancer and precancerous lesion.Methods: CD147, MMP-9 and TIMP-1 were detected by immunohistochemical stain in 49 cases of CIN(13 cases of CINⅠ,18 cases of CINⅡand 18 cases of CINⅢ),16 cases of squamou-cell carcinoma of the uterine cervix and 17 cases of normal cervical epithelial tissue. The data were analyzed statistically.Results: (1) In normal cervical epithelial tissue , CINⅠ~Ⅲand the squamou-cell carcinoma of the uterine cervix groups, the positive rates of CD147 were 11.76%(2/17),46.15%(6/13),66.67% (12/18), 88.89%(16/18) and 87.5% (14/16), respectively. The rates in CINⅠ~Ⅲgroups and invasive cervical carcinoma group were significant higher than that in normal cervical epithelial group (P<0.05).Combined with expression intensity, there was significant difference among the 5 groups of cervical tissue(χ2=33.93,P<0.001).To compare two groups randomly, there was significant difference, except CINⅠand CINⅡ, CINⅢand invasive carcinoma. To analyze the CINⅠ~Ⅲexclusively, the expression of CD147 were significant different among those three groups (P=0.002).(2) In normal cervical epithelial tissue , CINⅠ~Ⅲand squamou-cell carcinoma of uterine cervix, the positive rates of MMP-9 were 52.94%(9/17),84.62%(11/13),100%(18/18),94.44%(17/18)and 87.5%(14/16), respectively. Compared with CINⅡand CINⅢ, the positive rate of normal epithelium was significant lower(P<0.05). Combined with the expression intensity, there was significant difference among the 5 groups (χ2=32.41,P<0.001). To compare two groups randomly, there was significant difference, except CINⅢand invasive carcinoma . To analyze the CINⅠ~Ⅲexclusively, the expression of MMP-9 was significant different among those three groups (P<0.001).(3) In normal cervical epithelial tissue , CINⅠ~Ⅲand the squamou-cell carcinoma of the uterine cervix, the positive rates of TIMP-1 were 64.71% ( 11/17 ) ,84.62%(11/13),83.33%(15/18),77.78%(14/18) and 100%(16/16), respectively. The expression rates in CIN and invasive carcinoma were higher than normal epithelium, but in CIN, from CINⅠto CINⅢ, the expression rates had decrease tendency, and the tendency was not significant different in. Combined of expression intensity, there was significant difference among the 5 groups of cervical tissue(χ2=18.32,P=0.0011). To compare two groups randomly, the normal epithelium was significant different with other 4 groups ( P=0.011,0.006,0.005 and<0.001, respectively). To analyze the CINⅠ~Ⅲgroups exclusively, there are no difference.(4) In normal cervical epithelial tissue , CINⅠ~Ⅲand the squamou-cell carcinoma of the uterine cervix, the ratio that the MMP-9 expression intensity higher than that of the TIMP-1 were 11.26% (2/17),42.86%(16/49),56.25%(9/16),and there was significant different among the 5 groups(P=0.022). (5) In 5 groups, the expression of CD147 and MMP-9 was positive correlated(r=0.493,P<0.001). The similar results were found in CINⅠ,CINⅡand CINⅢ(r=0.362,P=0.011. In 5 groups, the expression of CD147 and TIMP-1 was positive correlated(r=0.369,P=0.001). To analyze the CINⅠ~Ⅲgroups exclusively, there are no difference. In 5 groups, the expression of MMP-9 and TIMP-1 was positive correlated(r=0.448,P<0.001). To analyze the CINⅠ~Ⅲgroups exclusively, there are no difference. Conclusions The expression rates and intensities of CD147 and MMP-9 are correlated with cervical lesion degree. The expression of CD147 and MMP-9 in different groups and in CINⅠ~Ⅲgroups were both positive correlated. The ratio of MMP-9/TIMP-1 is correlated with the cervical change stage. CD147 could be a good molecule marker. The expression rate of MMP-9 in normal cervical epithelial tissue is high, so it couldn't be an ideal tumor marker.
