Coronary heart disease is one of the common clinic aliments and treated as the mostthreaten to human health. Danhong injection is an effective Chinese herbal medicine used forclinic treatment of cardiovascular and cerebrovascular diseases. In this paper, according to theprescription of Danhong injection, Compound danhong pills made of Danshen, Honghua andbomeol was prepared by modern formulation technology. This medicine can be used forprevention and treatment of cardiovascular and cerebrovascular diseases as oraladministration.In this paper, the preparation recipe and method of Compound danhong pills wasscreened and optimized. The ethanol solution hot reflux extraction and ethyl acetateabstraction of the active ingredients of Danshen was firstly employed; Honghua activeingredients were prepared also by methanol solution hot reflux; The Danshen extract,Honghua extract and borneol were mixed with molten PEG4000 or PEG6000, then droppedinto the cooling agent consists of petronol and dimethyl silicone oil to get the dropping pills.The quantitative analysis method of the major ingredients in Compound danhong pillswere established and validated. Salvianolic acid B, tanshinoneâ…¡A and hydroxysafflor yellowA were analyzed by HPLC; Borneol was determined by GC and safflor yellows by UVspectrophotometric method. The results of method validation indicated: Each of the linearrelationship was good. The RSD of precision and repeatability were no less than 3% and all ofthe additive recoveries were between 95-105%.The material ingredients and the influence factor of Compound danhong pills werediscussed. 2-dimension fingerprint produced by HPLC-DAD was established. 10 commonpeaks of the chromatogram were identified by HPLC-MS, which revealed that the Compounddanhong pills were composed of danshen phenolic acids, tanshinones and safflor yellows. Inthe preparing procedure, salvianolic acid B, tanshinoneâ…¡A and borneol were stable, whilehydroxysafflor yellow A was unstable under this conditon (pH 3.3). A neol-keto equilibriumamong hydroxysafflor yellow A, iso-hydroxysafflor yellow A and safflor yellow A was foundfor the first time, which was influenced by the temperature and pH of the system.
