Of all among mechanisms, the bioactivity membrane lead to lipidperoxidation harm is induct the liver function crock up of important mechanism,protect the liver cell and anti-lipid peroxidation harm, To raise liver cell to bear anoxia to be subjected to dint and increase a liver storage, to surgical operation ofsuccess and recover from illness to have important and clinical meaning.This explore aim: We use Sixty-seven percent hepatectomy was performed inrat, To explore the effects of selenium enriched Spirulina platensis (Se-SP) onantioxidation and regeneration of rat hepatocytes after partial hepatectomy.Subject and methodsThe rat model of 67%partial hepatectomy (PH) in Wistar ratswas established.The ratswere rondomly divided into five groups: three operation group swereadministered intragastrically (i. g) every daywith150 mg/kg (H), 100 mg/kg (M) and15 mg/kg (L) of Se-SP according to bodyweight for seven days, two control groupsof placebo-control (P) and sham operation group (S) were treated with normalsaline (0.9%NaCl, NS). Then selenium (Se), glutathione peroxidase(GSHPx),superoxide dismutase (SOD), malondiaoldehyde (MDA) levels in hepatocytes weredetermined for different sampling time of 0 h, 24h respectively. The expression indexof proliferating cell nuclear antigen (PCNA) in hepatocytes was detected byimmunohistochemistry, and the DNA oxidation damage induced by peroxide wasanalyzed by comet assay.ResultsIn addition to group S, the concentrations of Se before the operation in the othergroups are significantly higher than before. The group before and after surgery theconcentration of MDA in rat liver cells were significantly different. In the two timepoints of after operation, the concentration of MDA in rat liver cells in Group H andM was significantly lower than the group N. In addition to the group S, The activityof GSHPx have significantly difference in before and after surgery, and The lattersignificantly higher than the former. The activity of SOD increased significantly aftersurgery. Furthermore, a high positive correlation (r=0.833.P=0.010<0.05) between Se level and GSHPx activity and a negative correlation (r=-0.878,P=0.004<0.05)between Se and MDA were found.The content of Se, MDA and the activity of SOD and GSHPx have increasedthan before the surgery, in which H and L group indices were significantly higher inall groups. In addition to the content of Se, the other indicators also increasedsignificantly in group C. The content of Se and the activity of GSHPx and SOD inGroup H was significantly higher than group C and S, However, MDA wassignificantly lower than that in group C. Hepatocyte PCNA index increasedsignificantly, hydrogen peroxide-induced DNA damage lesser extent.ConclusionsSe-SP performed well in antioxidation to hepatocytes in rat hepatectomy, Se-SPhas definite antioxidation and regeneration enhancing effects on rat hepatocytes afterhepatectomy, and suggesting high bioavailability of Se and up-regulation ofselenoenzymes expression might be responsible for these effects.
