Objective: To investigate the association of the serum concentration and single nucleotide polymorphism in matrix metalloproteinases(MMP)-1 and MMP-9 promoter with susceptibility to chronic venous insufficiency (CVI).Methods: Concentration of serum MMP-1 and MMP-9 were measured with Enzyme-linked Immunosorbent Assay.The genotype of MMP-1 and MMP-9 gene promoter region was determined by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP) .Results: The genotype frequencies of MMP-1 1G/ 2G in chronic venous insufficiency group were similar to those in control subjects. The frequency of the CC genotype in the MMP-9 gene polymorphism was significantly increased in CVI group when compared with controls (χ2 = 30.4,P < 0. 01). serum MMP-9 levels in CVI patients were significantly higher than those in healthy people. The expression of MMP-9 was closely related to clinical stages ( P < 0. 05)Conclusion:Individuals carrying MMP-9 C/C could significantly increase the risk of chronic venous insufficiency (CVI) but MMP一1 promoter SNP was not associated with the risk of chronic venous insufficiency (CVI).The serum levels of MMP-1 are no association with the risk of CVI. The higher serum levels of MMP-1 are strong association with the risk of CVI and clinical stages.
