Background:Neurotransmitters play important roles in secondary injury of nervous system.Glutamate is the most important excitatory neurotransmitters in the brain.Following traumatic brain injury(TBI),high concentrations of extracellular glutamate result in excitotoxic injury,as well brain microcirculation disturbance in addition to mechanical agents coup injury brain tissue,they concomitantly participated pathophysiological process after head injury.Excitatory amino acids(EAA)-glutamate excitotoxicity antagonists and free radical antagonists have largely researched,however how to reduce detriment product and promote elimination was less studied in the past.Objective:To set up an experimental animal model of traumatic brain injury to study the mechanism and the role of clearance therapeusis treatment TBI earlier.Methods:The traumatic brain injury model of rabbits was by utilizing a free-falling weight impactor device.Rabbits were randomly divided into 4 groups(n=10 in each group):sham operation control group,injury control group, treatment group,treatment group containing 2 subsets.Histological observation for each group after 6h TBI was carried.The water content of brain tissue was detected by dry-humid weight method.The contents of Na+ and Ca2+were detected by ICP methods.The contents of lactic acid were detected by automatic biochemistry analyzer.The contents of glutamate were detected by automatic amino acid analyzer.Results:The contents of brain tissue water,LACT,GLU,Na+ and Ca2+was markedly higher than that the control(p<0.01).Fitness degrade capillary hydrostatic pressure and a great quantity fluid to go out and come in can markedly reduce the contents of brain tissue water,LACT,GLU,Na+ and Ca2+, after traumatic brain injury(p<0.05).The treatments with mannitol,multiplicity Sodium Chloride,frusemide associated high dose Human serum albumin have favourable outcomes than that with mannitol,multiplicity Sodium Chloride and frusemide(p<0.05).Conclusion:The contents of brain tissue water,LACT,GLU,Na+ and Ca2+ markedly increased after traumatic brain injury,clearance therapeusis reduce the contents of brain tissue water,LACT,GLU,Na+ and Ca2+,after traumatic brain injury.Which indicate clearance therapeusis was efficacious.The possible mechanism of clearance therapeusis treatment TBI decrease detriment produce and promote elimination.
