Objective:To research the protective effects of phosphate and lecithin on Vero cells exposured NaAsO2.Methods:After processed cell by different dosage of NaAsO2,cells were cultured by nutrient fluid,which were added different dosage of trisodium phosphate,sodium hypophosphite and lecithin.We observed the effect of Vero cells multipli- cation-inhibiting through MTT assays,vitamin E as the standard control.The activity of Na+,K+-ATPase in Vero cells membrane were measured with spectrophotometric.The contents of membrane phospholipids in Vero cells were detected with HPLC.The micros- construction of Vero cells were observed with transmission electron microscope.Results:1)0.80~51.20mg/L NaAsO2had the inhibitory action to the Vero cells multiplication;three different dosage(69.00 mg/L,34.50mg/L,17.25mg/L) of trisodium phosphate and two kinds dosage of lecithin(17.77mg/L,8.88mg/L)had the protective functions to the cell contaminated by arsenic,and resisted the cells multiplication-inhibiting action induced by arsenic.Otherwise,the function of sodium hypophosphite(12.03 mg/L,24.07mg/L)was opposite,which had increased the toxicity of arsenic;2)2.20mg/L NaAsO2 inhibited the activity of Na+,K+ATPase in Vero cells membrane,compared with the control group,the high dosage trisodium phosphate and lecithin increased the activity of Na+,K+ATPase inhibited by NaAsO2(P<0.05),while the high dosage group sodium hypophosphite(24.07mg/L)was opposite function;3)The contents of four main component of membrane phospholipids(PC,PE,PS,SM)of Vero cells were decreased by NaAsO2(2.20mg/L).Trisodium phosphate(69.00mg/L)and lecithin(17.77mg/L)increased the contents of membrane phospholipids,which were decreased by NaAsO2(P<0.05);Conclusions:The protections of the trisodium phosphate, sodium hypophosphite and lecithin to Vero cells contamined by arsenic were bilateral such as multiplication-inhibiting action,activity of Na+,K+-ATPase and the contents of membrane phospholipids,which were related to the different states of phosphorus and the metabolism of different compound in vivo 69.00mg/L trisodium phosphate and 17.77mg/L lecithin interventional groups could protect cell exposed by NaAsO2 (2.20mg/L).
