Objectives: To evaluate the effects of PEDF eyedrops on the corneal allografe rejection by topical installition on the rats model of penetrating keratoplasy, and to investigate its possible mechansism and its correlation factor.Methods: Penetrating keratoplasty(PKP)was proformed orthotopically from wistar rats to SD rat recipients.60 SD rats were randomly assigned to two groups-experimental group and experimental control group after PKP;30 SD rats were proformed auto penetrating keratoplasty group(blank control group) . The experimental group was treated with PEDF eyedrops, the blank control group and experimental control group were treated with escipient eyedrops. All animals'surrival time was recored and compared by the biomicroscope examination. 6 animals in three groups were killed respectively at the 1st week, 2nd week, 3rd week, 4th week and 5th week postoperatively. CNV and inflammation were evaluated with HE staining. Corneal ultrastuctural morphology was observed by TEM. PEDF and VEGF expression was examined by immunohistochemistry. Results:1 Express level of VEGF of experimental control group is higher than of experimental group ( P<0.05 or P<0.01).2 The mean survival time of the allografts in the experimental group was 22.2±1.97days,The mean survival time of the allografts in the blank control group was 25.2±18.9days,but the mean surival time of the experimental control group was 11.1±1.25days, which was obviously longed as compared with the experimental control group (P<0.01).3 Express level of PEDF of experimental group is higher than of blank control group (P<0.05 or P<0.01) ; Express level of PEDF of experimental group is higher than of experiment control group (P<0.05 or P<0.01).Conclusion:1. PEDF eyedrops can depress express of VEGF in corneal allografts.2. Topical application of PEDF eyedrops can prolong suivival time of the allografts.3. PEDF eyedrops can enhance express of PEDF in corneal allografts.
