Objective: To explore the expression of survivin in testicular tissue of experimental varicocele rats and its correlation with spermatogenesis.Methods: 1. Experimental varicocele was induced by partial ligation of the left renal vein in wistar rats. 2. RT-PCR and immunohistochemisrty S-P method to detected: (1) survivin mRNA expression in testicular tissue of experimental varicocele rats. (2) survivin protein expression in testicular tissue of experimental varicocele rats.Results: 1. Spermatogenesis was effected by varicocele, and the damage get worse with varicocele time prolonged﹙P<0.05﹚. 2. Survivin mRNA and protein was expressed in testis of normal spermatogenesis rats. 3. The expression levels of survivin mRNA and protein was correlation with varicocele time; varicocele time longer, the expression of survivin was less in rats testic﹙P<0.05﹚. Survivin protein was mostly expressed in secondary spermatocytes and spermatoblasts, slightly expressed in leydig cells. 4. The expression levels of survivin mRNA and protein was related with pathological grade. A weakly expression of survivin mRNA was seen in specimens without haploid germ cells (pre-meiotic maturation arrest). But in specimens post-meiotic arrest, the expression levels seems no significantly difference with normal spermatogenesis.Conclusions: 1. Varicocele can result in spermatogenesis disorder, and there is significant correlation with survivin. 2. The expression levels of survivin is correlation with maintaining spermiogonium function, it suggest survivin maybe a regulatory gene in male gametogenesis.
