Objective To investigate the protective effects of injection ofCarthamus tinctorius on acute myocardial ischemia and its mechanismsby detecting S-T stages and T waves in rats' electrocardiogram(ECG),concentration of IL-6 and TNF-αin serum, Expression of bcl-2and bax, and index of haemorheology, and so on.Methods The rat model of acute myocardial ischemia (AMI) wasestablished with isoprenaline hydrochloride (ISO). Record everygroup of rats' ECG about S-T stages and T waves swing; concentrationof IL-6 and TNF-αin serum were checked by deploying balancingmethod and liquid phase competition method; immunohistochemistryand image analysis were carried out to investigate the changes ofexpressive intensity in apoptosis related Bcl-2 and Bax protein; therat model of blood stasis was established with adrenalinhydrochloride(Adr), and index of haemorheology in every group ofrats was detected.Result 1. After treated with Carthamus tinctorius and XiangDan, thedescended of the raise degree of S-T stages and T waves were odserved,the raise degree of S-T stages and T waves were obviously descended (P<0.05) at 1 hour and 48 hours ofter establishing model of AMI. Therewas not statistical significance between high dose group andXiangDan group(P>0.05), and compared with negative control groupthere was statistical significance (P<0.05); There was notstatistical significance between low dose group and negative controlgroup(P>0.05); 2. The concentration of IL-6 and TNF-αin serum ofall treated groups were decrease significantly. Compared withnegative control group, it is statistical significance(P<0.05);compared with w normal control group, it is not statisticalsignificance(P>0.05); 3. The expression of Bcl-2 and Bax proteinin all treat groups and negative control group increased, and ishigher than normal control; compared with negative control group,the Bcl-2 (OD) in high and low dose group(136.1±6.187,118.4±6.426)were significantly increased (p<0.05) and the Bax (OD)(76.7±4.880, 89.4±6.426)were significantly decreased (p<0.05);4. The blood stasis rats' whole blood viscocity obviously degraded,in high dose group, and it is statistical significance(P<0.05)compared with which of negative control group. All treated groupshave no obvious change in blood plasma viscocity, volume of packedred cells(VPRC) and blood sedimentation rate(BSR).Conclusion The injection of Carthamus tinctorius has effect ondegrading the raise degree of S-T stages and T waves of ischemia ECGand improving the function of ischemia myocard; effect onrestraining inflammatory reaction and the damage on myocard frominflammatory cell factors; The injection of Carthamus tinctorius canreinforce expression of Bcl-2 and refrain expression of Bax; candecrease blood plasma viscocity effectly. To summarize, theinjection of Carthamus tinctorius has function of protective and therapeutical effect on rats with acute myocardial ischemia.
